Advanced nanotherapeutics and targeted drug delivery for autoimmune and autoinflammatory disorders

Autoimmune and autoinflammatory disorders, including rheumatoid arthritis, psoriasis, inflammatory bowel disease, lupus, and multiple sclerosis, affect millions OF PEOPLES worldwide and are associated with chronic inflammation, immune dysregulation, and progressive tissue damage. Although biologics, corticosteroids, and disease-modifying drugs have improved disease management, long-term therapy is often limited by systemic toxicity, poor bioavailability, frequent dosing, loss of response, and patient non-compliance. Recent advances in nanotechnology and biomaterials have created new opportunities for more precise and effective therapies. Nanocarriers such as liposomes, polymeric nanoparticles, micelles, hydrogels, microneedles, and extracellular vesicle-inspired systems can enhance drug stability, improve targeting to inflamed tissues, and modulate immune pathways with reduced off-target effects. These innovations have the potential to transform treatment strategies for chronic immune-mediated diseases. This Research Topic aims to bring together cutting-edge research and expert perspectives on emerging nanotherapeutic and targeted delivery approaches for autoimmune and autoinflammatory disorders. The collection will address current challenges in conventional therapies, including limited tissue specificity, adverse effects, therapeutic resistance, and poor patient adherence. We seek to highlight innovative strategies that improve therapeutic outcomes through smart biomaterials, stimuli-responsive systems, localized delivery platforms, and precision immunomodulation. The Topic also aims to bridge fundamental immunology with translational pharmaceutical sciences by encouraging studies on disease mechanisms, immune–material interactions, preclinical efficacy, safety, and clinical potential. Contributions exploring personalized medicine, combination therapies, biologic delivery, gene/RNA-based therapeutics, and next-generation formulations are particularly encouraged. By integrating multidisciplinary advances, this collection will provide a valuable platform for accelerating safer, more effective, and patient-friendly interventions for immune-mediated inflammatory diseases. We welcome Original Research, Review Articles, Mini Reviews, Systematic Reviews, Methods, Perspectives, and Clinical/Translational studies related to autoimmune and autoinflammatory diseases. Topics of interest include, but are not limited to:• Nanocarriers for autoimmune disease therapy• Targeted delivery systems for rheumatoid arthritis, psoriasis, lupus, IBD, and multiple sclerosis• Liposomes, ethosomes, micelles, dendrimers, hydrogels, and microneedles• Biomaterials for immune modulation and inflammation control• RNA, gene, peptide, and biologic delivery platforms• Stimuli-responsive and precision medicine approaches• Drug repurposing through advanced delivery systems• Safety, toxicity, pharmacokinetics, and regulatory considerations• Clinical translation of nanomedicine for immunological disordersSubmissions integrating immunology, materials science, pharmaceutics, and translational medicine are highly encouraged.
