Advancements in Polymer- and Lipid-Based Nanoparticles for Enhancing Drug Solubility, Stability, and Bioavailability

Nanotechnology has significantly transformed pharmaceutical sciences by introducing nanomedicines that utilize nanoparticles to address traditional challenges in drug delivery. Polymer and lipid-based nanoparticles, such as nanoemulsions, liposomes, solid lipid nanoparticles (SLNs), and nanostructured lipid carriers (NLCs), are particularly prominent. Their biocompatibility and biodegradability, combined with their ability to safely transport a range of therapeutic agents, make these nanocarriers promising candidates for enhancing drug solubility, stability, absorption, and bioavailability.Despite their vast potential, these nanocarriers face substantial translational barriers. Variations between animal models and human physiology have posed challenges to clinical success, and the regulatory approval rate for new nanomedicines is still relatively low. However, successes in clinically approved nanoparticle-based formulations, examples include Doxil and Abraxane, highlight the potential for overcoming these barriers. Current research efforts are therefore dedicated to not only optimizing these drug delivery systems but also enhancing our understanding of their pharmacokinetics, biodistribution, and immune responses to better tailor nanomedicine to treat many diseases. To advance the field of nanomedicine, this Research Topic welcomes Original Research and Review contributions that explore issues in, but not limited to, the following areas:•     Design, synthesis, and functionalization of polymer- and lipid-based nanoparticles for drug-delivery.•     Innovative application of these nanocarriers in targeting specific diseases and enhancing therapeutic outcomes.•     Studies on the pharmacokinetics, drug transport, and systemic distribution of nanoparticle-based systems, with a focus on immune responses' long-term safety.•     Comprehensive reviews of the progress and challenges in drug delivery using polymer- and lipid-based nanoparticles, emphasizing translational barriers and regulatory aspects.
