Advances in Cytokine- and Cytokine-receptor-targeted Therapeutics
Since the introduction of infliximab for treatment of Crohn’s disease in 1998 (USA) and rheumatoid arthritis in 1999 (UK), there has been a major shift in approaches to management of inflammatory conditions and cancer as more and more biological therapies have been developed to target specific molecules involved in pathological processes. Ranging from neutralising monoclonal antibodies (mAbs) against immunoglobulins, through soluble cytokine receptor fusion proteins and growth factor receptor-linked kinase inhibitors, to mAbs targeting common chains of multiple cytokine receptors, new therapies have been devised to exploit multiple steps in the elicitation of cellular responses by extracellular signalling molecules.Over a quarter of a century on, our understanding of the roles of cytokines in pathologically altered physiological processes – and the application of targeted therapies to control them – continues to develop rapidly. This Research Topic will collect current research on cytokine- and cytokine receptor-targeted agents, their use to define the pathological roles of cytokines and their current and potential use to manage diseases that present unmet or incompletely met clinical needs. The goal of this topic is to present some important current developments in the understanding of the roles of cytokines and cytokine receptors in disease, and the current and potential use of therapeutic agents that target cytokines and their receptors. Contributions will address disease-specific roles of cytokines and their receptors, roles of cytokines in co-ordination/regulation of systemic inflammation, metabolism and tumorigenesis, and approaches to engineering of therapeutic agents to affect novel mechanisms of control of cytokine/receptor actions.Contributions are invited across all aspects of cytokine biology as related to disease – particularly inflammatory and metabolic conditions and cancer – and therapeutics. Articles focusing on the following themes are particularly welcomed:·development and application of bispecific monoclonal antibodies (mAbs) and mAbs targeting common chains of multiple receptors·roles of cytokines in metabolic conditions (diabetes, obesity, etc.) and potential for therapeutic intervention win anti-cytokine/receptor agents·co-ordinating roles of cytokines in local and systemic inflammation·recent developments in the roles of cytokines in musculoskeletal conditions and application of cytokine-targeted agents in their management·recent developments in the roles of cytokines in dermatological conditions and application of cytokine-targeted agents in their management·recent developments in the roles of cytokines in gastrointestinal conditions and application of cytokine-targeted agents in their management·recent developments in the roles of cytokines in respiratory conditions and application of cytokine-targeted agents in their managementTopic Editor Gordon Dent declares a financial interest in a company whose p38 MAPK inhibitor is the subject of research within this collection.
