Base and Prime Editing for Correction of Cancer-Associated Mutations

Recent advances in genome-editing technologies have unlocked the potential to directly correct pathogenic genetic mutations. Among these, base editing and prime editing represent innovative and highly precise approaches that hold particular promise for the correction of cancer-associated mutations. Unlike traditional genome-editing systems, base editors enable programmable, single-nucleotide conversion without introducing double-strand breaks, while prime editors expand the possibilities by enabling targeted insertions, deletions, and diverse base substitutions. This Research Topic aims to gather and highlight recent progress in the development and application of base and prime editing tools for the targeted correction of somatic mutations implicated in various cancers. We welcome original research articles, reviews, and perspectives that address but are not limited to:Development of new base and prime editors with improved specificity, efficiency, and versatility for cancer gene correction.Preclinical or in vitro studies demonstrating the correction of cancer-driver mutations using base or prime editing in relevant cancer models.Integration of base or prime editing with delivery platforms engineered for solid tumors or hematological malignancies.Comparative studies evaluating the safety, off-target effects, and therapeutic potential of base and prime editing versus other gene editing modalities in oncology.Challenges and opportunities for clinical translation of these gene editing approaches in cancer therapy, including ethical, regulatory, and technical considerations.By consolidating current efforts and insights, this Research Topic aims to provide a comprehensive overview of how base and prime editing can advance our ability to precisely correct cancer-associated genetic mutations, thus opening new horizons for precision oncology.
