Beyond Classical Monoamine Receptors: Emerging Regulators and Therapeutic Targets in Neuropharmacology
Monoaminergic neurotransmission has traditionally been understood through the pharmacology of dopamine, serotonin, and adrenergic receptors. However, growing evidence indicates that monoaminergic function is shaped by a broader network of receptors, transporters, intracellular signaling pathways, metabolic enzymes, and interactions with other neurotransmitter systems. Emerging targets, including trace amine-associated receptors (TAARs) are revealing previously unrecognized mechanisms regulating neuronal activity, behavior, and therapeutic responses. This Research Topic aims to bring together experimental, translational, and clinical research examining emerging mechanisms of monoaminergic regulation and their therapeutic relevance. Attention will be given to underexplored receptors, receptor–transporter interactions, intracellular and biased signaling, neurotransmitter-system crosstalk, and novel pharmacological approaches for neuropsychiatric, neurological, addictive, pain-related, and metabolic disorders. We welcome Original Research, Reviews, Systematic Reviews, Mini Reviews, Perspectives, and methodological studies addressing topics including, but not limited to:• Trace amines, TAAR1, and other trace amine-associated receptors• Emerging modulators of dopaminergic, serotonergic, and adrenergic transmission• Monoamine transporters and transporter–receptor interactions• GPCR signaling, receptor heteromers, and biased agonism• Monoamine metabolism and monoamine oxidase-related mechanisms• Crosstalk between monoaminergic, glutamatergic, GABAergic, opioid, and endocannabinoid systems• Novel pharmacological targets in addiction, psychosis, depression, chronic pain, cognitive impairment, and neurodegeneration• Translational biomarkers, imaging, medicinal chemistry, and drug discovery Topic editors: Evgeny Budygin, Raul R.
