Cancer Therapy Resistance: Mechanisms and Precision Strategies Targeting Ferroptosis, Oxidative Stress, Metabolic Reprogramming, and Immune Escape

Therapeutic resistance remains one of the most critical barriers to effective cancer treatment, limiting the long-term benefits of chemotherapy, radiotherapy, targeted therapy, and immunotherapy. Increasing evidence shows that resistance is driven by a complex interplay of intrinsic tumor cell programs and extrinsic factors within the tumor ecosystem, including oxidative stress, metabolic reprogramming, immune escape, and microenvironmental remodeling. Understanding and exploiting these interconnected mechanisms is central to developing next-generation precision oncology strategies. This Research Topic brings together basic, translational, and clinical work focused on the mechanistic underpinnings of therapy resistance and practical strategies to overcome it. A particular focus is placed on ferroptosis — a form of regulated cell death tied to lipid peroxidation and redox imbalance — which has emerged as both a resistance mechanism and a potential therapeutic vulnerability across multiple cancer types. Alongside this, we are interested in how metabolic reprogramming shapes treatment sensitivity, how the immune microenvironment enables or prevents effective therapy, and how biomarker-driven approaches can better match patients to treatments that work.We welcome submissions on, but are not limited to, the following themes:o Molecular and clinical mechanisms of resistance to chemotherapy, targeted therapy, radiotherapy, and immunotherapyo Ferroptosis and its regulation in the context of cancer treatment response and resistanceo Oxidative stress, redox homeostasis, and lipid peroxidation in therapy resistanceo Metabolic reprogramming — including mitochondrial function, lipid, amino acid, and glucose metabolism — as a driver of or target in resistanceo Tumor microenvironment remodeling and stromal contributions to therapy resistanceo Immune evasion and strategies to restore sensitivity to immunotherapyo Crosstalk between ferroptosis, metabolism, and immune regulationo Biomarker identification and validation for therapy resistance in clinical trialso Multi-omics approaches for mechanistic discovery and patient stratificationo Liquid biopsy and longitudinal monitoring of resistance evolutiono Rational combination strategies targeting resistance mechanisms based on molecular profiling Topic editors: Yinghao Cao, Zhuolun Sun, Jinxiao Li Source: https://www.frontiersin.org/research-topics/80182/cancer-therapy-resistance-mechanisms-and-precision-strategies-targeting-ferroptosis-oxidative-stress-metabolic-reprogramming-and-immune-escape
