Cell Death Regulation and Therapeutic Opportunities in Sepsis–Volume II

This Research Topic is the second volume in the series: Cell Death Regulation and Therapeutic Opportunities in SepsisThe previous volume can be viewed here: Volume I: Volume IThe field of sepsis research has advanced significantly in recent years, particularly regarding the interplay between immune responses and multiple forms of programmed cell death. Sepsis, now recognized as a leading cause of mortality in intensive care units worldwide, results from a dysregulated host response to infection that leads to widespread tissue damage and organ dysfunction. Despite progress in understanding inflammation and immune dysregulation in sepsis, current interventions remain insufficient, largely due to the complex and varied cell death mechanisms underlying the condition. Recent studies highlight the roles of apoptosis, necroptosis, pyroptosis, ferroptosis, and other cell death modalities, which not only compromise immune function but also propagate organ injury through the release of damage-associated molecular patterns (DAMPs). While substantial evidence links these processes to sepsis progression, critical gaps persist in our mechanistic understanding, especially regarding how cell death forms interact and how their products influence inflammatory and immune cascades. This Research Topic aims to systematically elucidate the relationships between programmed cell death and sepsis, with a specific focus on molecular regulatory mechanisms and their implications for diagnosis, prognosis, and therapy. Key objectives include deciphering the signaling crosstalk between various cell death pathways, understanding the timing and interplay of DAMP release, and identifying potential intervention points that could disrupt the cycle of inflammation and organ dysfunction in sepsis. By fostering multidisciplinary approaches and encouraging contributions that span basic science, biomarker discovery, and translational therapeutics, this topic seeks to advance new hypotheses and actionable strategies for improving sepsis outcomes. The scope of this Research Topic encompasses mechanistic, diagnostic, and therapeutic studies at the intersection of cell death and sepsis, without extending to unrelated systemic inflammatory disorders or non-infectious organ dysfunctions. To gather further insights within these boundaries, we welcome articles addressing, but not limited to, the following themes:o Mechanisms of various types of cell death and their interactions in sepsis.o Cell death-related signaling pathways and their modulation in sepsis pathogenesis.o Functions and dynamics of DAMPs in sepsis inflammation and immune regulation.o Identification and application of cell death-related biomarkers for sepsis diagnosis and prognosis assessment.o Development of novel therapeutic strategies targeting cell death pathways, including small molecules, antibodies, and nanobodies.o The influence of energy metabolism and metabolic reprogramming on cell death and sepsis progression.o Epigenetic regulation of aberrant cell death processes in sepsis.
