Cellular and Immune-Based Therapies in Lymphoid Malignancies
Lymphoid malignancies encompass a wide spectrum of hematologic disorders — including B-cell and T-cell lymphomas, Hodgkin lymphoma, and other indolent and aggressive subtypes — each with distinct biology, disease course, and response to treatment. While small-molecule targeted agents have transformed certain areas of this field, a large proportion of patients still face relapse or refractory disease, highlighting an urgent need for strategies that harness the immune system to produce deeper and more durable remissions.Over the past decade, cellular and immune-based therapies have moved from experimental approaches to established treatment options. CAR T-cell therapy has achieved unprecedented response rates in relapsed/refractory large B-cell lymphoma and follicular lymphoma; bispecific antibodies are reshaping frontline and salvage strategies across multiple subtypes; and allogeneic stem cell transplantation continues to offer curative potential for carefully selected patients. Emerging platforms — including NK cell therapy, TIL-based approaches, and next-generation CAR constructs — further expand the horizon. Yet significant challenges persist: immune effector cell-associated toxicities, primary and acquired resistance, manufacturing and access barriers, and uncertainty around optimal patient selection and treatment sequencing. This Research Topic focuses specifically on the cellular and immunological dimensions of lymphoid malignancy treatment, a complementary perspective to small-molecule and MRD-driven approaches. It aims to consolidate translational and clinical evidence on immune-based strategies, identify remaining gaps, and guide the rational integration of these therapies into routine practice. We welcome original research, reviews, mini-reviews, case reports, perspectives, and clinical trial reports addressing, but not limited to:-CAR T-cell therapy across lymphoma subtypes: efficacy, toxicity, and resistance-Bispecific antibodies in B-cell and T-cell lymphomas-Allogeneic stem cell transplantation: indications, conditioning, and graft-versus-lymphoma effects-Antibody–drug conjugates as immune-directed strategies in lymphoid malignancies-NK cell therapy, TIL therapy, and next-generation cellular platforms-Tumor microenvironment and immune evasion mechanisms in lymphoid cancers-Biomarkers of response and resistance specific to cellular and immune therapies-Combination strategies: cellular therapies with checkpoint inhibitors or other immunomodulatory agents-Real-world outcomes, access, and health disparities in cellular therapy deliveryTopic Editor Chijioke Nze received financial support for research from Daiichi Sankyo, BeOne therapeutics, Leuko Pointcheck, and Bristol Myers Squibb foundation.
