Chronobiology and Chronomedicine in the Third Millennium

Chronobiology is a dynamic field investigating the intricate timing mechanisms that govern physiological and behavioral rhythms. Emerging evidence from the past three decades demonstrates that circadian rhythms are fundamental not only to normal biological cycles but also to the development and progression of a wide array of diseases, including metabolic disorders, inflammatory syndromes, neurodegenerative conditions, neoplastic pathologies, and the aging process. The landmark Nobel Prize awarded in 2017 highlighted the centrality of molecular circadian mechanisms, further establishing their significance and catalyzing continued research. Nonetheless, translating this foundational knowledge into tangible improvements in clinical and therapeutic outcomes remains an ongoing challenge, calling for a paradigm shift in integrating chronobiology into medical research and practice. This Research Topic aims to foster a comprehensive understanding of how circadian rhythms influence disease mechanisms and therapeutic outcomes by promoting the adoption of chronobiological principles in both research and clinical care. Special emphasis is placed on the importance of precisely timed interventions—considering drug kinetics, the temporal structure of human physiology, and the circadian specificity of pharmacological targets—to optimize patient outcomes. Addressing these objectives involves elucidating the cellular and systemic pathways by which circadian disruptions contribute to disease and assessing innovative approaches to chronotherapy and chronomedicine. The scope of this Research Topic encompasses experimental, translational, and clinical studies that advance our understanding of circadian rhythm involvement in health and disease. We welcome contributions that delineate the mechanisms underlying circadian regulation, as well as those exploring the application of chronotherapy and chronomedicine across diverse medical domains. To gather further insights in the domains of chronobiology and its medical applications, we welcome articles addressing, but not limited to, the following themes:•The role of circadian rhythms in neurobiology and the pathogenesis of neurodegenerative diseases•Circadian regulation of metabolism and the implications for metabolic disorders•Chronobiological influences on cardiovascular and cerebrovascular physiology and diseases•The interplay between circadian mechanisms, carcinogenesis, and neoplastic disease progression•Development and evaluation of chronotherapeutic strategies for the prevention and treatment of disease•Articles encompassing reviews, original research, theoretical perspectives, methodologies, and translational studies are encouraged.
