Clinical and Investigational Biomarkers in Autoimmune and Autoinflammatory Diseases: From Classical Laboratory Medicine to Omics and Multiparametric Approaches.

Autoimmune and autoinflammatory diseases are complex, heterogeneous, and often difficult to diagnose and manage. Traditional laboratory diagnostics such as autoantibodies, complement, and acute-phase proteins have long formed the foundation of patient care, providing essential guidance for diagnosis, monitoring, and treatment. At the same time, advances in translational research are rapidly expanding the biomarker repertoire, offering new opportunities to refine disease understanding and improve outcomes. This Research Topic brings together original research, reviews, and expert perspectives on the evolving biomarker landscape in autoimmune and autoinflammatory diseases. A central theme is the complementary value of classical and modern approaches: established assays anchor clinical interpretation, while omics-based and multiparametric technologies provide deeper mechanistic insight and greater precision for patient stratification and therapy guidance. We are particularly interested in contributions highlighting knowledge and insights accumulated over years of clinical practice with established biomarkers, as well as those that explore how emerging technologies can build on and complement this foundation. The unifying goal is to show how diverse biomarker strategies can work synergistically — combining laboratory-based, mechanistic, and clinical perspectives — to advance understanding of disease activity and support therapeutic decision-making.Topics of interest include, but are not limited to:- Classical laboratory biomarkers (autoantibodies, complement, acute-phase proteins) in diagnosis and monitoring- Omics-based** and multiparametric approaches for mechanistic insight and patient stratification- Translational validation of novel biomarkers across the discovery-to-clinic continuum- Multi-analyte profiling and integrated analyses in autoimmune and autoinflammatory diseases- Biomarker strategies to guide therapeutic decision-making and therapy response monitoring- Synergistic use of established and emerging biomarker platforms in clinical practice*FAIR2 articles reporting omics datasets are particularly welcomed, ** Experimental validation (in vitro, in vivo or patient cohorts) of computationally- or omics-derived findings is expected Topic editors: Valentina Pucino, Onorina Berardicurti, Elena Pipi, Charikleia Kranidioti Source: https://www.frontiersin.org/research-topics/80190/clinical-and-investigational-biomarkers-in-autoimmune-and-autoinflammatory-diseases-from-classical-laboratory-medicine-to-omics-and-multiparametric-approaches
