Clinical Management in Huntington Disease: Experimental Therapies to Comprehensive Care

Huntington Disease (HD) represents one of the most complex and debilitating hereditary neurodegenerative disorders, profoundly affecting motor control, cognition, and mental health. It is caused by an expanded CAG trinucleotide repeat in the huntingtin gene (HTT), which leads to progressive neuronal dysfunction and death. Although significant progress has been made in understanding the genetic and molecular underpinnings of HD, the translation of these findings into effective disease-modifying therapies remains an unmet challenge. Current interventions are limited to symptomatic treatment, supportive care, and lifestyle adaptations aimed at preserving functional capacity. Recent studies have highlighted evolving approaches such as RNA-based therapies, gene-editing technologies, and modulators of protein homeostasis as promising, yet still experimental, avenues. However, gaps persist in the evidence base for long-term efficacy, standardization of clinical management across settings, and equitable access to emerging interventions, underscoring the need for integrated, patient-centered strategies. This Research Topic aims to advance knowledge and foster collaboration in the clinical management of Huntington Disease. By integrating multidisciplinary expertise, it seeks to consolidate contemporary clinical practices, explore innovative therapeutic developments, and identify barriers hindering optimal patient care. The overarching goal is to improve health outcomes and quality of life for individuals with HD through evidence-based, holistic, and forward-looking management frameworks. Specific objectives include assessing the current state of pharmacological and non-pharmacological interventions, evaluating predictive biomarkers for personalized care, and highlighting best practices that can be replicated across diverse clinical contexts. To gather further insights into the evolving landscape of Huntington Disease care, we welcome contributions addressing, but not limited to, the following themes:• Predictive and pharmacogenomic biomarkers• Modifier genes and their influence on disease variability• Clinical trials and emerging disease-modifying therapies• Pharmacological interventions and novel therapeutic targets• Symptomatic management of motor, cognitive, and psychiatric features• Multidisciplinary and integrated care models• The impact of digital and virtual care in neurodegenerative disorders• Mental health support and caregiver well-being• The role of patient support networks in rare neurodegenerative diseases Topic editors: Wai Lun Alan Fung, Erin Furr Stimming, Philippe Rizek, Zhuo Shao Source: https://www.frontiersin.org/research-topics/81103/clinical-management-in-huntington-disease-experimental-therapies-to-comprehensive-care
