Community Series in Genetic and Immunological Insights into Angioedema Without Wheals Volume II

This Research Topic is the second volume of the “Genetic and Immunological Insights into Angioedema Without Wheals ” Community Series. Please see Volume I here Hereditary angioedema (HAE) is traditionally defined as a bradykinin-mediated disorder caused by dysregulation of the kallikrein-kinin system. However, growing evidence suggests that the disease also has important immunological dimensions that remain insufficiently understood. In addition to abnormalities in vascular permeability, recent studies point to altered inflammatory biomarkers, endothelial activation, and interactions between immune and coagulation pathways, indicating that HAE is a more complex systemic disorder than previously appreciated. At the same time, the genetic basis of HAE continues to expand beyond SERPING1, with newly identified variants revealing further biological heterogeneity and leaving a significant proportion of patients with normal C1-inhibitor genetically unresolved. This Research Topic aims to advance understanding of the immunological and genetic mechanisms underlying HAE and their implications for diagnosis, disease classification, and patient care. We welcome original research, reviews, and clinically informative reports that explore the molecular and cellular immunopathogenesis of HAE, the identification of immune and genetic biomarkers, genotype-phenotype relationships, links between inflammation, endothelial dysfunction, and coagulation, and the immunological basis of comorbidities and disease variability. Contributions addressing emerging therapeutic strategies, including monoclonal antibodies, RNA-based treatments, and gene-editing approaches, are also encouraged where they provide insight into disease mechanisms or immunologically informed care. Through this collection, we seek to define HAE more clearly as an immune-relevant systemic disorder and support progress toward more precise diagnosis, improved stratification, and more effective, equitable management.Manuscripts without a focus on immunological mechanisms, such as those investigating other pathomechanisms of disease, are not in scope for Frontiers in ImmunologyTopic Editor Prof. Anastasios E Germenis is the co-founder of the spin-off company of the University of Thessaly CeMIA SA; The other Topic Editors declare no competing interests with regard to the Research Topic subject Topic editors: Matija Rijavec, Maurizio Margaglione, Anastasios E Germenis Source: https://www.frontiersin.org/research-topics/78594/community-series-in-genetic-and-immunological-insights-into-angioedema-without-wheals-volume-ii
