Community Series in Reducing Adverse Effects of Cancer Immunotherapy: Volume IV

This Research Topic is the fourth volume of the “Reducing Adverse Effects of Cancer Immunotherapy” Community Series. Please see Vol.I , Vol.II and Vol.III for further insights. What determines why some cancer patients endure harsher adverse effects from therapy than others? Is there an immunological key hidden within these differences? In the field of oncology, chemotherapy, radiotherapy, and more recently, immunotherapies, remain central in the fight against cancer, yet their use is often overshadowed by significant adverse effects such as fatigue, hair loss, severe nausea, and vulnerability to infections. These complications not only erode quality of life for patients, but also increase healthcare burdens and can force clinicians to reduce or interrupt cancer treatments.Notably, a deeper exploration into the underlying immunological mechanisms reveals that many patients must become immunocompromised to receive their therapy, which may predispose them to more severe toxicity and variable treatment responses, underscoring a pressing need to understand how immune status modulates these adverse outcomes. This Research Topic aims to inspire innovations that will alleviate and better manage the adverse effects of cancer therapies, with a focus on unravelling the immunological determinants that influence why certain individuals are more susceptible than others. By examining both the clinical presentation and the biological underpinnings of toxicity, especially as they relate to immune system manipulation, we seek to maintain treatment efficacy, improve tolerability, and prioritize patient well-being. Crucial questions remain around the specific pathways and immune responses involved, especially as therapies become more targeted and the cumulative impact of long-term immune modulation by new immunotherapies comes into sharper focus. This Research Topic focuses on advancing knowledge and management of side effects in cancer therapy, with particular emphasis on immunological factors and the distinct needs of immunocompromised patients. We welcome original research articles, reviews, mini-reviews, expert commentaries, and perspectives that address, but are not limited to, the following themes:- Immunological mechanisms underlying differential toxicity responses- Role of immune status and immunocompromised states in adverse effects- New pharmacological agents and immunomodulators aimed at side effect mitigation- Emerging immunotherapies and their long-term implications for immune system health- Innovative, technology-driven approaches to real-time toxicity assessment and management- Preventive strategies and supportive care for optimizing patient resilience during treatmentManuscripts consisting solely of bioinformatics or computational analysis of public genomic or transcriptomic databases without relevant clinical or biological validation are out of scope for this Research Topic.
