Current State and Innovative Strategies in Disease: Shared Mechanisms across Cardiovascular Disease, Inflammation, and Cancer

Cardiovascular disease and cancer are increasingly recognised as conditions that share pathophysiological ground rather than developing along separate tracks. Platelets sit at the centre of this overlap: beyond their role in thrombus formation, they interact directly with pathways of inflammation and immune response that also drive tumour progression and metastasis. Lipid and non-lipid mediators, including prostanoids and oxylipins, further link these two disease areas at the molecular level. Antiplatelet agents, aspirin foremost among them, now sit at the intersection of cardiovascular prevention and cancer chemoprevention, yet questions remain about who benefits, in which clinical contexts, and through which mechanisms. This Research Topic aims to consolidate current understanding of the mechanisms that connect platelet activation, inflammation, and immune signalling across cardiovascular disease and cancer, and to examine how this shared biology is being translated into therapeutic strategy. It brings together contributions addressing both the molecular and clinical dimensions of this overlap, from the regulation of platelet activation in health and disease to the efficacy, safety, and positioning of current and emerging antiplatelet agents. The Topic seeks to identify where mechanistic insight from one disease area can inform therapeutic or preventive strategy in the other, and to highlight open questions that warrant further investigation. This Research Topic follows the International Course on "Common Mechanisms in Cardiovascular Disease and Cancer: Focus on Platelet Activation and Inhibition" in Erice, Italy. Contributions are welcome from all researchers in the field, whether or not they attended. We welcome submissions addressing the shared mechanisms and therapeutic strategies linking cardiovascular disease and cancer, including but not limited to:• Mechanisms of platelet activation across different model systems and clinical settings• Interactions between platelets and inflammatory or immune response pathways in cancer and cardiovascular disease• Lipid and non-lipid mediators, including prostanoids and oxylipins, in cardiovascular disease and cancer• Efficacy and safety of current antiplatelet drugs, including aspirin, across diverse clinical contexts• Novel antiplatelet agents and emerging targets for platelet inhibition• Determinants of individual response to antiplatelet or chemopreventive therapyOriginal Research, Reviews, Mini Reviews, Perspectives, and Hypothesis and Theory articles are all welcome, provided they address platelet biology or the broader mechanistic and therapeutic links between cardiovascular disease, inflammation, and cancer.
