Deciphering the Human Glucolipid Metabolic Network: From Molecular Mechanisms to Precision Nutrition Strategies

Glucolipid metabolism is a core biological process for maintaining energy balance and homeostasis in the human body. However, the widespread adoption of high-energy-density diets and sedentary lifestyles has contributed to glucolipid metabolic disorders, which have become a global health challenge and represent a common pathological basis for obesity, type 2 diabetes, non-alcoholic fatty liver disease (NAFLD/MAFLD), and cardiovascular diseases. Although significant progress has been made over the past few decades in understanding individual metabolic pathways, glucolipid metabolism operates as a highly complex and interconnected network. The interconversion between carbohydrates and lipids, the intricate crosstalk among organs (e.g., the liver -muscle -adipose axis), and the dysregulation of nutrient -sensing pathways still require urgent in -depth exploration. Furthermore, inter-individual variations in genetic background, gut microbiota composition, and dietary patterns lead to heterogeneity in metabolic responses, limiting the effectiveness of traditional "one-size-fits-all" dietary recommendations. The goal of this Research Topic is to bring together cutting-edge research, integrating mechanistic insights at the molecular level with population-based nutritional interventions, thereby providing a theoretical foundation for precision nutrition strategies targeting the glucolipid metabolism. The Research Topic seeks to move beyond the traditional "one-size-fits-all" dietary advice by generating knowledge that supports personalized strategies based on an individual’s metabolism, genetic background, and gut microbiome. We invite contributions that explore the complex interplay between carbohydrate and lipid homeostasis, organ crosstalk (liver–muscle–adipose axis), circadian and gut microbiota influences, and novel dietary strategies. Studies employing multi-omics approaches (genomics, metabolomics, metagenomics) to characterize individual metabolic responses and to advance personalized nutritional interventions are particularly encouraged. This Research Topic focuses on the integrative regulation of human glucolipid metabolism—from molecular mechanisms to precision nutrition applications. Topics of interest include, but are not limited to:• Elucidating novel mechanisms underlying the interplay between glucose and lipid metabolism• Mechanisms of insulin resistance, lipotoxicity, mitochondrial dysfunction, and ER stress in metabolic disorders• Comprehensive investigation of organ crosstalk (liver–muscle–adipose–gut axis) and metabolic flexibility • Assessment of the efficacy of innovative dietary patterns (e.g., time-restricted eating, low-carbohydrate, low-fat, Mediterranean, plant-based) and their impact on glycolipid metabolism• Exploration of the role of gut microbiota–derived metabolites in host glucolipid metabolism and cardiometabolic health• Identification and validation of novel multi-omics biomarkers (using genomics, metabolomics, metagenomics, lipidomics) for early prediction, precision nutrition, and personalized intervention in metabolic diseases• Investigating the integrated regulation of metabolism by circadian rhythms and the gut microecology;• Promoting the identification of novel biomarkers using metabolomics, genomics, and other tools for the early prediction and personalized intervention of metabolic diseases.
