Deciphering the role of chronic inflammation in multi-systemic diseases and therapeutic strategies
Chronic inflammation has emerged as a critical factor in the pathophysiology of a broad spectrum of multi-systemic diseases. Increasing evidence indicates that persistent inflammatory processes are implicated in tumorigenesis, cardiovascular dysfunction, gynecological abnormalities, and gastrointestinal disorders, among others. Traditionally perceived as a localized protective mechanism, chronic inflammation is now recognized for its far-reaching effects that disrupt organ homeostasis and fuel disease progression. Current challenges include deciphering the specific molecular underpinnings that link inflammation with diverse clinical disorders, and understanding the dynamic interactions between systemic signals and local tissue environments. Although recent studies have shed light on key pathways such as NF-κB and NLRP3 inflammasome, as well as inflammatory cytokine networks, significant knowledge gaps persist regarding organ-specific mechanisms and cross-organ inflammatory crosstalk. This Research Topic aims to collect a multidisciplinary investigation into the cellular and molecular underpinnings of chronic inflammation across various organ systems. We seek to uncover novel inflammatory biomarkers, clarify the molecular transitions from acute to chronic inflammatory responses, and explore how targeted manipulation of inflammation can alter disease progression. By bringing together studies from basic immunology, translational research, and clinical practice, this collection aims to address unresolved questions, such as the mechanisms of inflammation-driven tissue remodeling and the efficacy of emergent anti-inflammatory interventions in reversing chronic disease. The scope of this Research Topic encompasses mechanistic, translational, and therapeutic dimensions of chronic inflammation in multi-organ disease contexts, while excluding studies limited solely to acute inflammation or non-inflammatory pathologies. We welcome submissions on—though not restricted to—the following research themes:• Molecular mechanisms of chronic inflammation in cancer progression, metastasis, and treatment resistance• Systemic inflammatory markers in cardiovascular and cerebrovascular disorders• Immune microenvironment and inflammatory drivers in gynecological diseases (e.g., endometriosis, PCOS)• Inflammatory signaling and gut-immune axis in gastrointestinal diseases• Novel therapeutic interventions targeting chronic inflammation and organ-specific sequelae• Discovery of diagnostic biomarkers for multi-systemic chronic inflammatory conditions• Interactions between metabolic dysfunction and chronic inflammation ("metainflammation”)We invite Original Research, Review, and Mini-Review articles addressing the above topics.
