Decoding Muscle–Bone Crosstalk in Health, Fracture Healing, and Repair
This Research Topic examines how muscle–bone communication governs skeletal integrity across health, injury, and disease. We welcome original research, reviews, and translational studies investigating the molecular, cellular, metabolic, and biomechanical mechanisms underlying bone-muscle crosstalk mechanisms during healthy growth, aging, adaptation to exercise and mechanical loading, unloading, and obesity, as well as in spinal cord injury and cancer (e.g., cachexia, metastases), neuromuscular pathologies (e.g., Duchenne muscular dystrophy), and treatment-related toxicities (e.g., chemotherapy systemic effects, activin/myostatin antagonism, lipid‑mediator‑based precision monitoring). Areas of interest include:• Lineage and fate decisions; • Mechanotransduction and mechano adaptive signaling; • Metabolic and redox control in muscle and bone (e.g., AMPK/mTOR, mitochondrial dynamics);• Osteoimmune signaling (TGF-β, TNF/IL-6, RANKL/OPG);• Myokine/osteokine interactions and impact on the musculoskeletal system;• The transition between osteochondral regeneration, fibrosis, and adipogenesis; • Lipid-Signaling Mediators;• Treatment-related toxicities (GLP 1 therapies, chemotherapy, etc.).Methodologies include single-cell and spatial profiling, lineage tracing, advanced imaging (MRI, micro-CT), bioengineering, biomaterials, rehabilitation protocols, and translational strategies (cell/EV therapies, anti-fibrotic approaches, and pro-regenerative cues). By bridging fundamental physiology with tissue repair and disease biology, this Research Topic aims to identify actionable mechanisms and therapeutic targets to preserve and restore coordinated muscle–bone and bone-muscle function across the lifespan.
