Emerging Druggable Targets and Chemoprevention Strategies in Solid Tumors

Solid tumors account for the vast majority of cancer-related morbidity and mortality worldwide. Despite substantial progress in surgical interventions, radiotherapy, and systemic therapies, treatment failure remains common due to intrinsic or acquired drug resistance, tumor heterogeneity, and late-stage detection. Consequently, there is an urgent need to shift cancer management toward two complementary fronts: intercepting malignant transformation through targeted pharmacological interventions, and identifying previously unexplored molecular vulnerabilities in established tumors.Recent breakthroughs in high-throughput screening, functional genomics, and structural biology have accelerated the discovery of novel druggable targets—including non-traditional kinases, chromatin remodelers, and metabolic regulators. Concurrently, advances in understanding oncogenic signaling pathways provide actionable opportunities to pharmacologically disrupt tumor progression before invasive disease emerges. Integrating precision interception with innovative targeted therapies represents a promising paradigm for durable cancer control. This Research Topic explores the pharmacology of emerging druggable targets and early pharmacological intervention strategies in solid tumors. Advances in target validation, molecular profiling, and drug design are opening new avenues for both arresting tumor progression and developing potent therapeutic agents for established disease. We welcome contributions on novel druggable targets and their mechanisms of action, targeted therapeutic agents and their molecular pathways, biomarker-driven target validation, preclinical and translational studies in solid tumor models, and strategies to overcome drug resistance.Topics of interest include:• Identification and validation of novel druggable targets• Signaling pathways, epigenetic regulators, and molecular mechanisms in solid tumor progression• Pharmacological cancer interception: mechanism-based small molecules and targeted modulators• Biomarkers for target selection and therapeutic efficacy• Preclinical models and translational studies demonstrating cancer-cell selectivity• Combination strategies integrating targeted therapy and pharmacological interception• Drug resistance and strategies to overcome it in solid tumors Topic editors: Surya P Singh, Agnieszka Zdzisława Robaszkiewicz, Chujiao Hu Source: https://www.frontiersin.org/research-topics/85976/emerging-druggable-targets-and-chemoprevention-strategies-in-solid-tumors
