Endocannabinoid and Lipid Signalling in Brain Health and Disease: From Glia and Neurons to Neuroinflammation and Tumours
The endocannabinoid system (ECS) is one of the most widely distributed lipid signalling networks in the central nervous system, regulating synaptic plasticity, glial activation, immune tone and cell survival. It operates alongside a broad bioactive lipidome, including lysophospholipids, sphingolipids and specialised pro-resolving mediators, which collectively shape cellular responses in health and disease. Accordingly, disruption of these neurolipid signalling networks is increasingly recognised across a range of brain disorders. Altered endocannabinoid and/or lipid signalling contributes to oxidative stress and synaptic failure, fine-tunes glial cell metabolism and function to amplify or resolve inflammatory cascades, and modulates proliferation, autophagy and immune evasion in primary brain tumours.Despite this breadth, key mechanistic questions remain open. Cell-type-specific roles of ECS components in astrocytes, oligodendrocytes microglia and neurons in rodent and human disease models are not yet resolved, the contribution of lipid mediators beyond canonical endocannabinoids to brain (dys)function is underexplored, and translational connections to biomarkers and therapeutic targets remain limited. This Research Topic aims to consolidate mechanistic and translational research on endocannabinoid and lipid signalling in brain health and disease, with a focus on how these molecular pathways govern glial and neuronal responses and how they are reprogrammed in neuroinflammation and brain tumours. A longer-term goal is to connect these insights to biomarker discovery and molecularly informed therapeutic strategies. We welcome Original Research, Review, Mini Review, Methods, Hypothesis and Theory, Perspective and FAIR2 articles on the following sub-topics:o ECS components, including CB1, CB2 and GPR55, in glial and neuronal signalling under physiological conditions.o Bioactive lipid mediators, including lysophospholipids, sphingolipids, and eicosanoids, in brain function.o ECS and lipid signalling in neurodegenerative diseases, inflammatory disorders and pain.o Bioenergetic metabolism of astrocytes, oligodendrocytes and microglia, and its modulation by endocannabinoids and lipid mediators in brain pathology.o ECS and lipid signalling in glioma and glioblastoma.o Pharmacological modulation of the ECS and related lipid pathways as a therapeutic strategy in neuroinflammation and tumours: from preclinical models to translational evidence.o Lipidomic approaches for biomarker discovery in brain disease, including biofluid and human tissue studies.Topic Editor Antonio Zottola provides a consultancy service to Aelis Farma related to the measurement of cellular respiration regulated by endocannabinoids, and will not handle any related manuscripts.
