Endocrine Disruptors and Synaptic Plasticity: Molecular Pathways and Regulatory Challenges
Endocrine disruptors have gained significant attention for their ability to interfere with neurodevelopment and adult brain function, most notably by altering synaptic plasticity. Compounds such as diethyl phthalate (DEP) and perfluorooctanesulfonic acid (PFOS) have been implicated in disrupting neuronal signaling, synaptic strength, and plasticity through diverse molecular pathways. Recent research highlights their influence on neuronal proliferation, apoptosis, and differentiation, mediated in part by the dysregulation of microRNAs and central signaling cascades including the Ras and PI3K/Akt/mTOR pathways. Despite mounting evidence linking these chemicals to adverse neural outcomes, ongoing debates remain regarding permissible exposure limits and appropriate regulatory oversight to safeguard public health. These uncertainties underscore the need for comprehensive evaluation of both mechanistic effects and policy frameworks. This Research Topic seeks to advance our understanding of how exposure to endocrine disruptors affects synaptic plasticity at the molecular and cellular levels. The goal is to clarify the role of specific signaling pathways and miRNA regulation in mediating neurotoxic effects, and to identify vulnerable stages of neurodevelopment or brain regions. Additionally, the Topic aims to gather critical perspectives on regulatory challenges, risk assessment strategies, and the translation of mechanistic findings into actionable guidelines or therapeutic interventions. Unraveling these processes is essential to protect neural health and inform future regulatory standards. We welcome submissions including original research, reviews, perspectives, and regulatory analyses focusing on the intersection of endocrine disruptors, synaptic function, and policy. Relevant themes include, but are not limited to:· Regulation of synaptic plasticity by endocrine disruptors· MicroRNA-mediated effects on neuronal proliferation and apoptosis· Disruption of Ras and PI3K/Akt/mTOR signaling in neural circuits· Identification of neurodevelopmental windows of vulnerability· Regulatory standards, challenges, and therapeutic interventions for neurotoxic exposures We encourage a wide range of submissions, including original research, methods papers, brief research reports, data reports, systematic reviews, mini reviews and perspectives.
