Engineering Brain Therapeutics: Biomaterials and Precision Delivery from Disease Mechanism to Translation
Neurological disorders remain difficult to treat because biological barriers, complex disease microenvironments and limited regional specificity restrict how well therapeutics reach and act on their targets in the brain. At the same time, the molecular and biophysical features of the diseased brain, from barrier breakdown and inflammation to changes in tissue stiffness and matrix composition, offer new opportunities to design therapies that respond to the disease itself. This Research Topic brings together engineered and biomaterial platforms that turn mechanistic understanding of brain disease into precise and effective treatment, across neurodegeneration, traumatic brain injury, stroke and brain tumours. We welcome contributions that keep a clear link between disease mechanism and therapeutic design, including but not limited to:• Strategies for crossing biological barriers, including the blood brain barrier, and achieving region specific delivery• Biomaterial platforms such as peptide hydrogels, nanoparticles, exosomes and engineered carriers• Nose to brain and other minimally invasive routes of administration• Controlled distribution and release tuned to the disease microenvironment and its underlying mechanisms• Therapeutic approaches that respond to or modulate the biophysical properties of the diseased brain, such as matrix mechanics and mechanotransduction, with a focus on how physical and electrical cues are sensed and transduced at the molecular and cellular level• Translational proof of concept in vivo, from bioengineering to therapeutic effect in specific brain diseases• Approaches that connect mechanistic discovery with therapeutic design and deliveryWe welcome Original Research, Reviews, Mini Reviews, Methods, Perspectives and Brief Research Reports.
