Evolving paradigms: IL-5's role in eosinophilic pathologies and targeted treatments

The field of cytokine biology has seen significant advancements, particularly concerning Interleukin-5 (IL-5) and its role in eosinophil-related disorders. IL-5 is largely responsible for the expansion and activation of eosinophils, which contribute to pathologies such as eosinophilic asthma, eosinophilic granulomatosis with polyangiitis (EGPA), hypereosinophilic syndrome (HES), and eosinophilic gastrointestinal diseases. Despite the availability of anti-IL-5 therapies like mepolizumab, reslizumab, and benralizumab, there are still challenges to address. There exist ongoing debates over optimal approaches to patient selection, the resistance mechanisms encountered by these therapies, biomarkers for predicting response, and the assessment of long-term safety and efficacy. This Research Topic aims to illuminate recent advancements and ongoing challenges in the therapeutic targeting of IL-5 in eosinophilic pathologies. By investigating cutting-edge research, reviewing past developments, and offering translational insights, we seek to refine the current understanding and future potential of IL-5 targeting strategies. The primary focus lies on expanding the evidence base of IL-5's role beyond traditional eosinophilic diseases to include areas like cancer immunology, chronic rhinosinusitis with nasal polyps, and atopic dermatitis. The scope of the research is comprehensive yet focused on enlightenment of IL-5 biologics and their potential. To gather further insights in IL-5-targeted studies, we welcome articles addressing, but not limited to, the following themes:-Fundamental mechanisms of IL-5 signaling and eosinophil regulation-Biomarkers for diagnosis and prognosis related to IL-5 targeting-Advancements with IL-5 receptor signaling and eosinophil heterogeneity-Interactions of IL-5 with other cytokine networks in tissue inflammation-Novel therapeutic applications of IL-5 modulation beyond eosinophilic pathologiesWe seek diverse article types such as original research, reviews, systematic reviews, clinical trials, case series, and perspectives, emphasizing scientific rigor and clinical applicability. Studies that explore treatment variability, resistance pathways, or next-generation biologics will be prioritized. Complete with discussions on implications for practice and potential new therapies, this Research Topic aims to accelerate the adoption of IL-5-directed therapies and inspire innovations in managing eosinophilic diseases.
