Exploring GPCR Dynamics: Advanced Bioanalytical Techniques for Membrane Protein Study and Characterization

In cellular biology, G Protein-Coupled Receptors (GPCRs) play a pivotal role in signal transduction, comprising the largest family of eukaryotic membrane proteins. They serve as essential mediators of physiologic responses and are at the forefront of therapeutic targeting due to their involvement in numerous physiological processes. However, elucidating the precise mechanisms by which ligand binding leads to structural changes in GPCRs, which subsequently determine signaling specificity and efficacy, remains challenging due to their inherently dynamic and flexible nature, compounded by their integration into the cell membrane. The recent surge in technological advancements has markedly improved our ability to investigate GPCRs, with tools such as single-molecule FRET (smFRET), cryo-electron microscopy (cryo-EM), nuclear magnetic resonance (NMR), hydrogen-deuterium exchange mass spectrometry (HDX-MS), double electron-electron resonance (DEER), and molecular dynamics (MD) simulations unveiling unprecedented insights. Additionally, advancements in bioanalytical instrumentation, including surface plasmon resonance (SPR), microscale thermophoresis (MST), mass photometry, and biolayer interferometry (BLI), allow researchers to precisely analyze GPCR interaction kinetics and related structural transitions. This Research Topic aims to consolidate cutting-edge research in GPCR pharmacology, structural biology, and systems biology, alongside technological innovations that foster novel scientific discoveries. It endeavors to enhance the understanding of GPCR conformational landscapes, interaction networks, and regulatory mechanisms within environments closely mimicking their native states. To gather further insights in the diverse methodologies and approaches in GPCR research, we welcome articles addressing, but not limited to, the following themes:- Molecular and kinetic characterization of GPCR structural transitions and interactions with ligands and signaling partners- Novel expression and purification platforms for isolating functional GPCRs and signaling partners- Innovative technologies for functional reconstitution of GPCRs in membrane-mimetic environments such as nanodiscs and liposomes- Advances in genetic code expansion for site-specific labeling of GPCRs for biophysical studies- Development of emerging biosensors and fluorescent probes for live-cell GPCR trackingWe invite original research articles, reviews, perspectives, and technical notes to contribute to this comprehensive exploration of GPCRs.
