Exploring the Pharmacological Modulation of Potassium Channels: Implications for Channelopathies and Therapeutic Innovations
Potassium channels are integral membrane proteins that play a crucial role in maintaining cellular homeostasis by regulating potassium ion flow across cell membranes. They are vital for a wide range of physiological processes, including neuronal excitability, muscle contraction, and cardiac rhythm. Dysfunction in potassium channel activity, often resulting from genetic mutations or regulatory disturbances, can lead to a group of disorders known as channelopathies. These include epilepsy, cardiac arrhythmias, and various neuromuscular diseases. Also, potassium channels also play emerging roles in immune function, cancer progression, and metabolic regulation, broadening their significance in health and disease especially in rare diseases.Recent advances in the understanding of potassium channel structure and function have opened new avenues for therapeutic interventions. Pharmacological modulation of these channels offers significant potential for the development of innovative treatments for channelopathies. This Research Topic aims to gather cutting-edge contributions that explore both the fundamental aspects and the translational potential of targeting potassium channels. We invite researchers to submit original research articles, reviews, and perspectives that cover, but are not limited to, the following themes: o Structural and Functional Insights: Investigating the molecular architecture of potassium channels and their regulation under physiological and pathological conditions. o Novel Modulatory Compounds: Discovery and characterization of small molecules, peptides, or biologics that can modulate potassium channel activity, with an emphasis on specificity and mechanism of action. o Pathophysiological Mechanisms: Elucidating how defects in potassium channels contribute to the pathogenesis of channelopathies. o Therapeutic Innovations: Preclinical and clinical studies demonstrating the application of potassium channel modulators in disease models. o Technological Advances: Development of new tools and techniques for studying potassium channels, including computational modeling, electrophysiology, and high-throughput screening methods. o Case Studies and Clinical Trials: Reports on clinical experiences with potassium channel-targeting drugs, highlighting successes, challenges, and future directions. Through this Research Topic, we aim to foster a multidisciplinary dialogue that bridges basic research and clinical application, ultimately contributing to the advancement of therapeutic strategies in the realm of potassium channelopathies.
