Exploring the role of NLRP3 inflammasome in chronic inflammation and associated diseases

The NOD-like receptor family pyrin domain containing 3 (NLRP3) inflammasome is a pivotal protein complex involved in the body's response to harmful stimuli, including foreign pathogens and self-danger signals. Its activation leads to the conversion of pro-caspase-1 into active caspase-1, which in turn promotes the secretion of inflammatory cytokines like interleukin 1β (IL-1β) and IL-18. These cytokines play crucial roles in mediating pro-inflammatory responses and initiating pyroptotic cell death. While inflammation is essential for defense, its chronic manifestation can contribute to various diseases such as cardiovascular ailments, diabetes, neurological disorders, autoimmune conditions, and certain cancers. Aberrant activation of the NLRP3 inflammasome is increasingly implicated as a central factor in the development and progression of these conditions, thereby making it a vital target for therapeutic interventions aimed at reducing chronic inflammation.Recent studies underscore the potential of inhibiting the NLRP3 inflammasome as a promising strategy for managing inflammation-related diseases. Despite significant scientific advances, gaps persist in understanding the precise mechanisms through which the NLRP3 inflammasome contributes to disease pathogenesis. These gaps necessitate comprehensive investigations to develop precision-targeted anti-inflammasome therapies. The purpose of this Research Topic is to provide insights into chronic inflammation and related diseases, with a particular focus on innovations targeting the NLRP3 inflammasome. This Research Topic specifically aims to illuminate how NLRP3 inflammasome activation functions and influences chronic inflammation and related diseases, encompassing cardiovascular diseases, neurological disorders, autoimmune diseases, and cancers. Additionally, it seeks to create a platform for discussions on recent developments in interventions, therapies, and diagnostics pertaining to the NLRP3 inflammasome in context with chronic inflammation and associated conditions. To gather further insights into this expansive area, we welcome articles addressing the following themes, including but not limited to:• The roles and mechanisms of NLRP3 inflammasome activation in related diseases, including cardiovascular, neurological, and cancerous conditions• Evolutionary mechanisms of chronic inflammation and its impact on related diseases• Interaction between the NLRP3 inflammasome and chronic inflammation, and their joint roles in disease• Research on drug development and inhibitors targeting the NLRP3 inflammasome as therapeutic approaches against inflammatory diseases• Additional advancements in NLRP3 inflammasome and chronic inflammation researchWe welcome a variety of manuscript types, including original research articles, reviews, and short communications related to these themes.
