Folate Metabolism, Oxidative Genomic Instability, and Predictive Modeling of Disease Progression in Hematologic Disorders

Folate-dependent one-carbon metabolism is central to nucleotide biosynthesis, methylation balance, and redox homeostasis. Disruption of this tightly regulated metabolic network may lead to homocysteine accumulation, oxidative stress, impaired DNA repair, and genomic instability. Increasing evidence suggests that polymorphisms in the methylenetetrahydrofolate reductase (MTHFR) gene and altered folate metabolism may create a permissive environment for oxidative DNA damage, particularly reflected by the accumulation of 8-oxo-2'-deoxyguanosine (8-oxoG).While individual components of folate metabolism and redox imbalance have been extensively studied, their integrated contribution to the pathogenesis, clinical heterogeneity, and progression of hematologic disorders remains incompletely understood. In both hereditary blood disorders such as thalassemia and hematologic malignancies including lymphomas, chronic oxidative stress and metabolic dysregulation may converge to drive genomic instability and influence disease course. This Research Topic seeks to establish a mechanistic framework connecting folate metabolism, oxidative DNA damage, and genomic instability in hematologic diseases. By integrating metabolic, genetic, redox, and computational perspectives, the aim is to identify molecular vulnerabilities, predictive biomarkers, and potential therapeutic targets. We welcome contributions addressing:• One-carbon metabolism and its impact on genomic stability• Functional and clinical consequences of MTHFR polymorphisms• 8-oxoG and related oxidative DNA lesions as biomarkers of disease activity• Crosstalk between folate metabolism, epigenetic regulation, and redox signaling• Mathematical and computational modeling approaches integrating metabolic, genetic, and oxidative biomarkers to predict disease severity, progression, and clinical outcomes• Translational and therapeutic strategies targeting metabolic-redox imbalanceOriginal research articles, comprehensive reviews, translational investigations, and predictive modeling studies are encouraged. Advancing mechanistic insight into folate-driven redox dysregulation, combined with integrative modeling approaches, may facilitate precision diagnostics and personalized therapeutic strategies in hematologic disorders.
