G-Proteins in Focus: From Molecular Mutations to Physiological Implications

G-proteins and G-protein coupled receptors (GPCRs) are central to signal transduction and are integral in regulating numerous physiological processes, including heart rate, blood pressure, neuronal activity, memory, cognition, and sensory perceptions such as vision and pain. With new evidence emerging about the role of G-proteins in diseases, there is a particular interest in their involvement in congenital pathologies. Recent advances in genetic studies, alongside the use of heterologous expression systems, single-molecule techniques, and in silico modeling, have opened avenues to study mutations' impact and their association with certain phenotypic abnormalities more deeply. Notably, mutations in subunits, such as Gβ1, responsible for complex syndromes like GNB1e, affecting intellectual development and motor functions, and Gαs, linked with MacCune-Albright syndrome, Gαq, associated with melanoma, and Gβ2, implicated in neurological disorders and sick sinus syndrome, are under active investigation. This Research Topic aims to explore the structural and functional implications of mutations within various G-proteins and their critical roles in both normal physiology and disease states. The main objectives include understanding the molecular mechanisms triggered by these mutations, identifying their effects on cellular function, and delineating how these changes manifest clinically. By addressing specific questions and testing hypotheses related to these genetic alterations, the goal is to uncover potential therapeutic targets and advance our knowledge of disease pathogenesis associated with G-protein mutations. To gather further insights within this scope of research, we welcome articles addressing, but not limited to, the following themes:Functional analysis of mutations in different G-protein subunits and their structural repercussionsRole of G-protein mutations in congenital and acquired abnormalitiesIn-depth studies on the impact of mutations on signal transduction pathwaysModelling studies to predict functional consequences of G-protein mutationsExploration of therapeutic strategies targeting G-protein mutations in disease contextsWe are inviting contributions in various formats, including original research papers, reviews, and case studies.
