Genetic Determinants of the Oral Microbiome–Bone Axis in Remodeling, Repair, and Disease
In recent years, significant advancements have been made in understanding how oral flora impacts not only bone health but also the complex interplay between local oral and systemic environments in bone remodeling and repair. This has catalyzed a broader appreciation for the multifaceted biochemical networks through which oral microbiota, together with host genetics, modulate skeletal physiology. Emerging research highlights how microbial metabolites, inflammatory mediators, and host immune responses collectively influence both the local microenvironment of alveolar bone and systemic bone metabolism and repair mechanisms throughout the body. The evolving landscape of bone homeostasis and repair is now recognized as being shaped by dynamic interactions between the oral microbiota and genetic predispositions. These factors contribute not only to conditions like osteoporosis and altered bone mineral density but also influence bone healing and regeneration following injury or disease.Understanding the mechanisms through which oral and systemic factors, together with genetic variability, impact bone structure and restoration is essential for advancing innovative therapeutic interventions and preventative strategies. This Research Topic aims to illuminate the molecular, cellular, and clinical pathways whereby oral flora, systemic processes, and genetic signals converge to modify bone health, remodeling, and repair. By thoroughly mapping these interactions, the research seeks to foster new scientific narratives capable of transforming current approaches to both the prevention and management of bone-related disorders. We welcome original research articles and reviews addressing, but not limited to, the following themes:• Studies elucidating the impact of oral flora on bone remodeling and repair via microbial metabolites, inflammatory pathways, and immune responses at both local (oral) and systemic levels.• Investigations into the synergistic and potentially antagonistic effects of oral microbiota, systemic factors (such as hormones or nutrition), and genetic determinants in shaping bone health, disease susceptibility, and the efficacy of bone healing.• Research focused on the contributions of oral and genetic factors to bone diseases (including osteoporosis, periodontitis, osteomyelitis), with an emphasis on innovative prevention and treatment strategies.• Contributions that offer novel insights into the mechanisms and interconnectivity between oral flora, systemic influences, and genetic factors in bone metabolism, regeneration, and disease, as well as proposals for future research directions.
