Genome-Engineered CAR-T and CAR-NK Cell Therapies for Hematological Malignancies.

Recent advances in genome editing technologies, such as CRISPR/Cas9 and TALENs, have revolutionized the field of adoptive cell therapies, offering innovative new strategies to enhance the efficacy and safety of CAR-T and CAR-NK cell therapies for hematological malignancies. These approaches make it possible to precisely engineer immune cells to improve tumor targeting, persistence, resistance to immunosuppression, and minimize adverse effects. Genome editing also opens up new avenues for generating allogeneic, off-the-shelf cellular therapies, broadening access for patients with blood cancers. This Research Topic aims to bring together original research articles, comprehensive reviews, perspectives, and clinical trial reports addressing recent advancements in genome-edited CAR-T and CAR-NK cell therapies for the treatment of leukemias, lymphomas, myelomas, and other hematological malignancies. Areas of interest include, but are not limited to:1. Innovative genome editing tools and strategies for CAR-T and CAR-NK engineering.2. Mechanisms underlying improved antitumor activity and persistence.3. Approaches to reduce immunotoxicity and off-tumor effects.4. Preclinical and clinical data on genome-edited CAR therapies for blood cancers.5. Overcoming challenges such as immune escape and tumor microenvironment resistance.6. Development and characterization of universal or off-the-shelf CAR-T/NK products.7. Safety, ethical, and regulatory considerations in genome-edited cell therapies.Contributions discussing translational aspects, novel delivery systems, and future directions in the field are highly encouraged. We welcome both basic and clinical research that will help shape the next generation of cellular immunotherapies for hematological malignancies.
