Genomics, Tumor Microenvironment, and Treatment Selection
The fields of cancer genomics and tumor microenvironment research have witnessed rapid advancements in recent years, fundamentally altering our understanding of cancer development, progression, and response to therapy. While genomic profiling has enabled the identification of key oncogenic drivers and actionable mutations, the role of the tumor microenvironment in shaping tumor behavior and therapeutic vulnerability is now recognized as equally pivotal. The intricate interactions between cancer cells, surrounding stromal and immune cells, and the extracellular matrix greatly influence tumor growth, metastasis, and resistance to treatment. Despite major progress, integrating these diverse layers of biological information to inform effective clinical decision-making remains an ongoing challenge.Recent studies have demonstrated that both genomic alterations and the composition of the tumor microenvironment can predict sensitivity or resistance to a range of targeted therapies, immunotherapies, and chemotherapies. For example, emerging evidence highlights how the presence of specific immune cell subsets or stromal factors can modulate the efficacy of treatments designed around genomic targets. Large-scale initiatives such as The Cancer Genome Atlas and single-cell sequencing efforts have further revealed the complexity and heterogeneity of tumors, illustrating the necessity of multidimensional profiling in precision oncology. This Research Topic aims to bridge the gap between genomic analysis and microenvironmental assessment by exploring how their integration can guide optimal treatment selection for cancer patients. Key objectives include elucidating the mechanisms by which tumor genomics and the microenvironment interact to influence therapy response, identifying new biomarkers for treatment stratification, and showcasing innovative methodologies - such as multi-omics approaches - for comprehensive tumor characterization. By fostering interdisciplinary dialogue, the Research Topic seeks to accelerate the translation of combined genomic and microenvironmental insights into clinical practice. To gather further insights in the integration of genomics and tumor microenvironment research for precise treatment selection, we welcome articles addressing, but not limited to, the following themes:• Mechanisms of interaction between tumor genetic alterations and the microenvironment• Predictive biomarkers and signatures arising from integrated genomic and microenvironment profiling• Advances in multi-omics and spatial analysis technologies for cancer characterization• Strategies for combining tumor genomics and microenvironment data in clinical decision-making algorithms• Case studies or clinical trials exemplifying personalized treatment approaches based on comprehensive tumor profiling• Challenges and opportunities in translating preclinical findings to patient care• Novel therapeutic interventions targeting both genetic and microenvironmental drivers of cancer Topic editors: Silvia R Rogatto, Wilfredo Alejandro González-Arriagada, Dimitris Tatsis Source: https://www.frontiersin.org/research-topics/78211/genomics-tumor-microenvironment-and-treatment-selection
