Harnessing Combination Therapies and Pharmacological Synergy Against Infectious Diseases: Innovative Approaches and Clinical Impact
The ongoing rise of antimicrobial resistance, complex co-infections, and persistent infections such as those involving biofilms have intensified the need to optimize infectious disease pharmacotherapy. Combination therapies—administering two or more antimicrobial agents—offer a promising approach to enhance efficacy, mitigate resistance, and overcome pharmacological limitations of monotherapies. However, rational drug combinations demand a deeper understanding of synergistic, additive, and antagonistic interactions, as well as a thorough evaluation of pharmacokinetic and pharmacodynamic profiles. This Research Topic welcomes original research, systematic reviews, and perspectives on the science and clinical application of combination therapies in infectious diseases. We welcome submissions addressing, but not limited to:• Novel methodologies to assess and predict pharmacological synergy or antagonism (in vitro, in vivo, in silico).• Clinical experiences and trial outcomes using combination therapy for bacterial, viral, fungal, or parasitic infections.• Mechanistic studies targeting biofilms, intracellular pathogens, or infections in specific populations (e.g., immunocompromised hosts, co-infected patients).• Drug–drug interactions: beneficial and adverse outcomes, with a focus on co-infection management (e.g., TB/HIV, malaria/HIV).• Strategies to overcome antimicrobial resistance using combination treatments.• Advances in drug delivery systems optimizing the effects of combination regimens.• Regulatory, pharmacoeconomic, and implementation perspectives of combination therapy in infectious disease care.By bringing together multidisciplinary research, this Topic aims to accelerate understanding and clinical translation of effective combination strategies, ultimately improving patient outcomes in the face of evolving infectious threats.
