Hormonal Regulation in Cancer: From Molecular Mechanisms to Precision Oncology
Hormonal regulation plays a pivotal role in the development, progression, and therapeutic response of numerous cancers, particularly breast, prostate, endometrial, ovarian, thyroid, and adrenal malignancies. Hormone receptors and endocrine signaling pathways regulate key cellular and molecular processes, including proliferation, differentiation, metabolism, survival, and genome stability. Over the past decade, advances in genomics, transcriptomics, epigenetics, and molecular profiling have substantially expanded our understanding of hormone-dependent tumor biology at the mechanistic level. Increasing evidence further demonstrates that hormonal signaling interacts with oncogenic pathways, the tumor microenvironment, immune system, and metabolic networks, thereby driving tumor evolution and therapeutic resistance.Despite major advances in endocrine therapies and targeted treatments, resistance to therapy remains a significant challenge across hormone-related cancers. Elucidating the molecular and cellular mechanisms underlying hormone-dependent tumorigenesis, disease progression, and treatment resistance is essential for improving patient outcomes. Recent developments in multi-omics technologies, functional perturbation approaches, liquid biopsy integrated with biological validation, and precision oncology frameworks have accelerated the identification of mechanistically relevant biomarkers and therapeutic targets. This Research Topic aims to bring together basic scientists, translational researchers, and clinicians to explore the complex role of hormonal regulation in cancer, with a strong emphasis on mechanistic and experimentally validated insights. We seek to foster interdisciplinary research that bridges fundamental molecular discoveries with biologically grounded clinical applications, highlighting innovative approaches for understanding tumor biology and improving therapeutic strategies. Particular emphasis will be placed on studies that advance mechanistic understanding of cancer evolution, endocrine resistance, and tumor plasticity. We welcome Original Research, Review, Mini Review, and Perspective articles on themes including, but not limited to:- Hormone receptor signaling in cancer- Molecular mechanisms of hormone-dependent tumorigenesis- Endocrine resistance and mechanistically defined therapeutic vulnerabilities- Genomic, epigenetic, and transcriptomic regulation of hormonal pathways- Mechanistically informed biomarkers for diagnosis, prognosis, and treatment response- Tumor microenvironment and immune-endocrine interactions with functional validation- Mechanistic insights into precision oncology and therapy response- Novel endocrine therapies and targeted treatments supported by molecular and cellular evidence- Liquid biopsy and molecular monitoring of hormone-driven tumor evolution- Translational and experimental studies in hormone-related cancersPlease note: manuscripts consisting solely of bioinformatics, computational analysis, or predictions of public databases which are not accompanied by validation (independent clinical or patient cohort, or biological validation in vitro or in vivo, which are not based on public databases) are not suitable for publication in this Research Topic.
