Immune Regulation and Therapeutic Response in Solid Tumors: Mechanisms, Modulation, and Translational Pathways
Treatment efficacy in solid tumors is profoundly influenced by immune regulation within the tumor microenvironment. The interplay between tumor and host immunity determines whether immunotherapies achieve durable responses or fail. Unlike isolated biomarker studies, understanding how regulatory circuits among T cells, macrophages, stromal cells, and cytokine networks shape cancer progression is key to refining therapeutic outcomes. This Research Topic seeks to integrate mechanistic and translational studies describing how immune regulation dictates therapy response in solid tumors. By bridging cellular immunology, translational oncology, and therapeutic optimization, it aims to clarify the pathways that determine responsiveness, resistance, or adverse immune reactions. Contributions merging basic mechanistic insight with clinical relevance, especially studies exploring regulatory checkpoints, immune homeostasis, or dynamic biomarkers, will be prioritized. To capture the diversity of current research, we welcome contributions including but not limited to:• Mechanisms of immune regulation and feedback within solid tumor microenvironments.• Crosstalk among immune cells, stromal elements, and tumor cells influencing treatment response.• Interventions that modulate immune balance to enhance checkpoint therapy efficacy.• Translational studies linking immune regulatory signatures to clinical outcomes.• Integrated multi-omics or spatial analyses uncovering regulatory patterns predicting efficacy. We invite researchers to contribute Original Research, Reviews, Mini Reviews, and Perspectives addressing how immune regulation can be leveraged to improve the success of current and next-generation therapies. Studies bridging mechanistic discovery and clinical application are particularly encouraged. Please note: manuscripts consisting solely of bioinformatics, computational analysis, or predictions of public databases which are not accompanied by validation (independent clinical or patient cohort, or biological validation in vitro or in vivo, which are not based on public databases) are not suitable for publication in this journal.
