Immune responses in liver disease and solid malignancies: mechanisms, transplantation, and clinical outcomes
The heterogeneity of the tumor immune microenvironment (TIME) and its surrounding architecture significantly impacts immune responses and the efficacy of immunotherapy in solid tumors. A key challenge in overcoming therapeutic resistance lies in decoding the regulatory mechanisms underlying patient- or population-specific immune subtypes, which could be leveraged to enhance immunotherapy or other treatments. When combined with surgical resection, innovative therapies that offer low carcinogenic potential and enhanced tissue repair capabilities may provide dual benefits for cancer treatment and tissue regeneration.Recent advances in methodologies, including new approach methodologies (NAMs), real-world studies, big data and multi-modal bioinformatics, tissue engineering, micro-nano scale therapies, and interdisciplinary approaches, are reshaping oncological and immunological research with future-oriented implications. Thus, innovative studies across oncological immunology and transplant immunology at all levels will drive personalized and precision clinical practice, ultimately improving survival and quality of life for patients with liver diseases and solid malignancies.By utilizing advanced technologies, NAMs, real word data, multi-modal bioinformatics and big data, bioengineering and interdisciplinary approaches, this issue aims to address the critical problem of varied drug response in cancer therapies and dysregulated immune response in transplantation.
