Immunogenetics of complex diseases: From genetic variants to immune dysregulation

The field of immunogenetics is pivotal in understanding the complex interplay between genetic predisposition and immune system dysregulation. This intricate relationship is central to a wide array of complex diseases, including autoimmune disorders, chronic inflammatory conditions, infectious diseases, and certain cancers. With the rapid advancements in high-throughput sequencing technologies, genome-wide association studies (GWAS), and systems immunology, researchers are uncovering a deeply complex immunogenetic landscape that governs disease susceptibility and pathophysiology. Despite these advances, there remain significant gaps in understanding the full scope of genetic contributions and their interactions with the immune system. This Research Topic aims to spotlight the latest discoveries in immunogenetics, emphasizing how genetic variants shape immune responses and contribute to the progression of complex diseases. We invite research articles that investigate genetic and epigenetic mechanisms, delve into gene-environment interactions, and explore innovative integrative multi-omics approaches. We particularly encourage studies that utilize bioinformatics, machine learning, and functional genomics to interpret immune-related genetic signals effectively. Through these efforts, we aim to create a comprehensive understanding of immunogenetic influences on disease. We welcome articles addressing, but not limited to, the following themes:• Defining the boundaries and limitations of systems immunology in understanding immunogenetics.• Identify novel immunogenetic markers and pathogenic pathways.• Exploring innovative therapeutic targets informed by systems immunology approaches in complex diseases.• Bridging the divide between computational systems discoveries and practical clinical applications.• Investigate gene-environment interactions and their influence on immune regulation. We welcome diverse article types such as original research, reviews, and perspectives that contribute to advancing the understanding of the immunogenetic framework in complex diseases.
