Immunogenic Consequences of Non-Apoptotic Regulated Cell Death Modalities: Necroptosis, Ferroptosis, Pyroptosis, Cuproptosis, and PANoptosis

The field of cancer biology has recently recognized the significance of immunogenic cell death (ICD), a process by which dying cells emit alarm signals—predominantly through the release of damage-associated molecular patterns (DAMPs)—that recruit and activate the immune system. With over twenty distinct forms of regulated cell death (RCD) now identified, attention has turned to several non-apoptotic RCD modalities, including necroptosis, ferroptosis, pyroptosis, cuproptosis, and PANoptosis, which have shown the capacity to elicit immunogenic effects. Recent studies indicate that pharmaceutical modulation of these cell death pathways holds promise for amplifying the efficacy of cancer immunotherapies. However, the immunological consequences of these RCDs are multifaceted, influencing tumor progression and treatment responsiveness in complex and sometimes contradictory ways. This Research Topic aims to advance understanding of the immunogenic effects produced by non-apoptotic RCD modalities, with an emphasis on recent discoveries, ongoing controversies, and prospective therapeutic implications. A key objective is to elucidate the novel signals and molecular mechanisms by which different cell death programs drive or inhibit antitumor immune responses. By fostering a rigorous, critical assessment of the pro- and anti-tumorigenic actions of these pathways, this Research Topic seeks to provide a balanced perspective on the feasibility and risks of targeting non-apoptotic RCDs for immunotherapeutic benefit. To gather further insights in the context of cancer and immunotherapy, the scope of this Research Topic is centered on, but not restricted to, the immunogenic impacts and pharmaceutical targeting of non-apoptotic RCDs. We welcome submissions addressing, but not limited to, the following themes:Molecular mechanisms underlying the immunogenic effects of necroptosis, ferroptosis, pyroptosis, cuproptosis, and PANoptosisSignaling pathways and DAMP release from dying cancer cellsApplication of RCD modulators for therapeutic purposes, particularly in cancer treatmentTargeting RCDs in both tumor cells and tumor microenvironment componentsDual anti- and pro-tumorigenic effects and challenges in pharmaceutical modulation of non-apoptotic RCDsAppendix:We welcome Original Research articles, Reviews, and Mini-reviews.
