Inflammasomes and Neuroimmune Interactions: Bridging Innate Immunity and Neurodegeneration

This Research Topic explores the role of inflammasomes as key regulators of neuroimmune interactions, linking innate immune activation to neurodegeneration. Inflammasomes, such as NLRP3, are cytosolic complexes that detect cellular stress and activate pro-inflammatory cytokines like IL-1β and IL-18, as well as pyroptotic cell death. While critical for CNS homeostasis, their chronic activation contributes to the progression of neurodegenerative disorders, including Alzheimer's, Parkinson's, multiple sclerosis, and ALS. This Research Topic addresses the urgent need to understand how dysregulated innate immune responses, particularly inflammasome activation, contribute to the onset and progression of neurodegenerative diseases. Inflammasomes, once considered restricted to pathogen defense, are now recognized as pivotal mediators of sterile neuroinflammation and drivers of chronic CNS pathology. Despite significant advancements in characterizing inflammasomes in peripheral tissues, their precise role in neuroimmune crosstalk, microglial priming, glial-neuronal interactions, and long-term neural dysfunction remains incompletely understood. By gathering cutting-edge research and expert reviews, this collection seeks to unify disparate findings into a cohesive narrative around inflammasome biology in the brain. The ultimate goal is to define key mechanistic links between inflammasome activity and neurodegeneration and to highlight promising therapeutic targets that modulate these pathways with precision. We welcome contributions that explore the role of inflammasomes in CNS pathology through the lens of neuroimmune interactions. Manuscripts may include, but are not limited to the following themes:• Molecular mechanisms of inflammasome activation in microglia, astrocytes, and neurons • Crosstalk between inflammasomes and other inflammatory pathways (e.g., TLRs, STING) • Inflammasome-driven cytokine signaling and its effects on neural circuits • Peripheral immune modulation of CNS inflammasomes • Therapeutic targeting of inflammasome components in neurodegenerative disease • Identification and validation of inflammasome-related biomarkers in CSF or blood We accept original research articles, systematic or narrative reviews, mini-reviews, perspectives, and hypothesis-driven opinion pieces. Interdisciplinary and translational studies, as well as those involving in vivo models and clinical correlates, are encouraged.
