Integrative Approaches in Neuroblastoma Research: Pathogenesis and Strategies for Diagnosis and Prognosis
Neuroblastoma (NB) is the most common pediatric solid tumor located extracranially, accounting for up to 8% of pediatric malignancies. The clinical course of NB is highly heterogeneous, ranging from spontaneous regression or differentiation to treatment-refractory progression despite intensive therapy. The survival rate for high-risk NB is less than 40% despite multimodal therapy, including surgery, highly intensive chemotherapy, radiation therapy, and immunotherapy. Pediatric patients with high-risk neuroblastoma have poor survival rates and urgently need more effective treatment options with fewer side effects. Neuroblastomas are infiltrated by natural killer (NK), T, and B cells, as well as immunosuppressive myeloid populations. NK cells show reduced cytotoxicity, and T cells have a dysfunctional profile. Tumors are complex ecosystems, defined by spatiotemporal interactions among heterogeneous cell types, including malignant, immune, and stromal cells. Research to unravel the intricacies of neuroblastoma development is pivotal to developing a management strategy. This Research Topic aims to provide compelling evidence and insights into the pathogenesis of neuroblastoma development, and develop diagnosis and prognosis strategies for neuroblastoma. By exploring these dimensions, the Research Topic endeavors to identify crucial knowledge gaps and advance the management spectrum of neuroblastoma. Efforts will focus on facilitating progressive strides in understanding the disease, thereby fostering the development of more innovative and effective management approaches. We welcome original research, review, clinical trial, and case report articles that address the following subtopics● Genomic studies of neuroblastoma● Immune microenvironment of neuroblastoma● Metastasis mechanisms of neuroblastoma● Tumor heterogeneity in neuroblastoma● Applications of artificial intelligence in the diagnosis of neuroblastoma● Clinical prognostic factorsPlease note: Manuscripts consisting solely of bioinformatics, computational analysis, or predictions of public databases which are not accompanied by validation (independent clinical or patient cohort, or biological validation in vitro or in vivo, which are not based on public databases) are not suitable for publication in this journal.
