Iron Dysregulation, Oxidative Stress, and Inflammation: Mechanisms and Therapeutic Insights Volume II
This Research topic is the second volume of the successful Research Topic “Iron Dysregulation, Oxidative Stress, and Inflammation: Mechanisms and Therapeutic Insights”. Please see the first Volume here.Iron homeostasis is essential for maintaining cellular and systemic health, as iron is required for a wide range of metabolic and enzymatic processes. However, disruption of iron uptake, storage, transport, or utilization can promote the accumulation of redox-active iron and excessive production of reactive oxygen species (ROS), thereby contributing to oxidative damage, inflammatory signaling, and tissue dysfunction. Increasing evidence indicates that these interconnected processes play a central role in the initiation and progression of diverse acute and chronic diseases.Recent advances have further highlighted the complex relationship between iron metabolism, oxidative stress, inflammation, and regulated forms of cell death. In particular, ferroptosis, an iron-dependent form of regulated cell death characterized by lipid peroxidation, has emerged as an important mechanism linking iron and redox biology to immune regulation and disease progression. The interplay between ferroptosis and other forms of regulated cell death, together with its connections to metabolic reprogramming, mitochondrial dysfunction, and immune responses, has opened new avenues for understanding disease mechanisms and identifying therapeutic targets. The clinical relevance of these interconnected pathways is increasingly recognized across a broad range of diseases. Cancer, neurodegenerative disorders such as Alzheimer’s and Parkinson’s diseases, and cardiovascular diseases are closely associated with dysregulation of iron metabolism, oxidative stress, and inflammatory signaling. Understanding how these processes interact in different disease contexts may provide new opportunities for identifying disease biomarkers, elucidating pathological mechanisms, and developing targeted therapeutic strategies. This Research Topic aims to provide an updated perspective on the molecular and cellular mechanisms underlying iron dysregulation and its interaction with oxidative stress and inflammation. We particularly welcome studies addressing emerging regulatory pathways, iron-dependent signaling mechanisms, ferroptosis and related forms of regulated cell death, as well as translational and therapeutic approaches targeting these interconnected processes. Topics of interest include, but are not limited to:• Iron homeostasis and inflammatory signaling: molecular mechanisms regulating iron uptake, storage, transport, and utilization, and interactions with inflammatory pathways in health and disease.• Iron-driven oxidative stress: mechanisms through which iron dysregulation and ROS contribute to cellular damage, metabolic dysfunction, and disease progression.• Ferroptosis and regulated cell death: molecular regulation of ferroptosis, lipid peroxidation, iron-dependent signaling, and interactions with other forms of regulated cell death.• Immunometabolism and iron biology: interactions between iron metabolism, immune cell function, inflammatory responses, and the tissue microenvironment.• Mitochondrial and metabolic mechanisms: the roles of mitochondrial dysfunction, metabolic reprogramming, and redox imbalance in iron-associated pathologies.• Disease mechanisms and biomarkers: the contribution of iron dysregulation, oxidative stress, inflammation, and ferroptosis to cancer, neurodegenerative, cardiovascular, metabolic, and other chronic or acute diseases, including the identification of potential biomarkers.• Therapeutic strategies: emerging pharmacological and molecular approaches targeting iron metabolism, oxidative stress, ferroptosis, inflammatory signaling, and related pathways.• Translational and clinical perspectives: preclinical and clinical studies evaluating therapeutic targets, diagnostic approaches, biomarkers, and strategies for the prevention or treatment of diseases associated with iron dysregulation. This Research Topic welcomes Original Research, Reviews, Mini-Reviews, Methods, Perspectives, and Technology and Code, and other article types that provide new mechanistic, methodological, or translational insights into the relationship between iron metabolism, oxidative stress, inflammation, and disease.
