Laryngeal Cancer: Integrative Approaches from Molecular Mechanisms to Clinical Management

Laryngeal cancer represents a significant challenge within the field of head and neck oncology, primarily due to its profound effects on critical functions such as speech, swallowing, and airway protection. Persistent difficulties exist in achieving optimal oncological outcomes while preserving laryngeal function, reflecting the heterogeneity observed both clinically and at a molecular level. Recent studies have highlighted the complex biological landscape of laryngeal cancer, including substantial molecular and microenvironmental diversity that underpins tumor progression, treatment resistance, and varied patient responses. While there is mounting evidence pointing to key molecular alterations and potential biomarkers, their uptake into clinical workflows remains slow. New advances in molecular profiling, imaging, immunotherapies, and minimally invasive interventions continue to expand the horizon of personalized care. Still, a critical gap exists in integrating these innovations seamlessly into routine decision-making and in ensuring improved patient-centered outcomes. This Research Topic aims to accelerate the translation of biological discoveries into clinical practice for laryngeal cancer, bridging the divide between laboratory research and real-world patient care. The main objective is to foster a multidisciplinary platform that unites basic science, translational, and clinical research to collectively address challenges such as therapy resistance, tumor heterogeneity, and the balancing of oncological control against preservation of function and quality of life. By promoting collaboration among laboratory scientists, clinicians, and allied health professionals, the Research Topic seeks to identify, validate, and implement clinically actionable biomarkers, refine patient stratification, and drive innovation in both surgical and non-surgical management, including emerging immunotherapies, targeted approaches, and rehabilitative strategies. This Research Topic encompasses the entire translational spectrum, but focuses on the integration of multidisciplinary research to enhance diagnosis, management, and post-treatment quality of life for patients with laryngeal cancer. Submissions may address any stage of disease or aspect of care, but should align with the translational and patient-centered ethos of this initiative. We welcome articles addressing, but not limited to, the following themes:- Molecular and microenvironmental mechanisms shaping carcinogenesis and treatment resistance.- Diagnostic, prognostic, and predictive biomarker discovery and validation.- Advances in targeted therapies, immunotherapy, and multimodal treatment strategies.- Surgical and non-surgical innovations, including minimally invasive and organ-preservation approaches.- Improvements in imaging, diagnostics, and intraoperative technologies.- Functional outcomes, quality of life, and rehabilitation (voice, swallowing, survivorship).- Interdisciplinary models for integrating laboratory discoveries into clinical management. Please note that manuscripts consisting solely of bioinformatics or computational analysis of public genomic or transcriptomic databases that are not accompanied by validation (independent cohort or biological validation in vitro or in vivo) are out of the scope of this Research Topic.
