Lipid Nanoparticles: A Historical Journey to Neurological Therapeutics

Lipid nanoparticles (LNPs) have emerged as a transformative platform in the field of neurotherapeutics, particularly for targeting the aging brain. Historically, the central nervous system has posed substantial obstacles to effective drug delivery due to the blood-brain barrier and the complexity of neural tissue. Over recent decades, research on LNPs has advanced rapidly, fueled by the need to improve the treatment of neurodegenerative disorders and age-related cognitive decline. Noteworthy preclinical and clinical studies have demonstrated the capacity of LNPs to encapsulate and deliver a range of molecular cargoes, from nucleic acids to small molecules, with increased specificity and reduced systemic toxicity. Despite these breakthroughs, challenges remain in optimizing LNP design for selective brain targeting, minimizing immunogenicity, and ensuring long-term safety, particularly in aging populations. This Research Topic aims to trace the evolution of lipid nanoparticles as a technology for neurological therapeutics, with a special focus on their application in brain aging. We seek to highlight both foundational developments and contemporary advances that have shaped the role of LNPs in treating neurodegenerative conditions. Key objectives include elucidating the mechanisms by which LNPs interact with neural cells, evaluating their efficacy in preclinical and clinical models, and identifying the hurdles that must be overcome to translate this technology into mainstream therapeutics for aging-related neurological disorders.Within the boundaries of cellular and molecular mechanisms of brain aging, we welcome studies that explore the historical progression, current applications, and future prospects for LNPs in neurotherapeutics. Submissions can address, but are not limited to, the following themes:o Historical milestones in the development of lipid nanoparticle technology for neurological applicationso Cellular and molecular mechanisms influencing LNP transport across the blood-brain barriero Advances in LNP engineering for targeted delivery of therapeutics in brain aging and neurodegenerationo Application of LNPs in gene editing and RNA-based treatments for age-related neurological diseaseso Safety, immunogenicity, and translational challenges in the use of LNPs for elderly populationso Comparative studies of LNP-based versus traditional drug delivery systems in neurological contextsWe encourage a wide range of submissions, including original research, methods papers, brief research reports, data reports, systematic reviews, mini reviews and perspectives.
