Mechanisms Linking Cancer and Metabolic Diseases: From Molecular Pathways to Therapeutic Interventions Volume II

This Research topic is the second volume of the successful Research Topic “Mechanisms Linking Cancer and Metabolic Diseases: From Molecular Pathways to Therapeutic Interventions”. Please see the first Volume here.Cancer and metabolic diseases, including obesity, diabetes, and metabolic syndrome, are increasingly recognized as interconnected conditions that share complex molecular, cellular, and systemic mechanisms. Metabolic reprogramming in cancer cells, chronic inflammation, insulin resistance, and oxidative stress are key factors linking these diseases. The Warburg effect, glutaminolysis, and altered fatty acid metabolism in cancer cells, together with the inflammatory and metabolic dysregulation associated with obesity and diabetes, can create a tumor-permissive microenvironment. Understanding these complex interactions is crucial for identifying novel molecular targets and developing effective prevention and therapeutic strategies for both cancer and metabolic disorders. The goal of this second volume is to further explore the mechanisms linking cancer and metabolic diseases and to highlight emerging discoveries, methodological advances, and therapeutic opportunities in this rapidly evolving field. We aim to bring together research investigating the bidirectional relationships between metabolic dysfunction and cancer, with particular emphasis on shared molecular pathways, metabolic heterogeneity, tumor–host interactions, and novel approaches for therapeutic intervention. This collection will provide a comprehensive overview of the current understanding of these interconnected conditions and offer insights into future research directions. This Research Topic welcomes contributions that explore the molecular and cellular mechanisms linking cancer and metabolic diseases, with a focus on shared pathways such as metabolic reprogramming, inflammation, insulin resistance, and oxidative stress. We particularly encourage submissions that investigate the role of lifestyle factors (e.g., diet, physical activity, and smoking) in the development and progression of both conditions and explore potential therapeutic interventions, including metabolic inhibitors, pharmacological treatments, and lifestyle modifications. Specific themes of interest include, but are not limited to:• Metabolic reprogramming in cancer (e.g., the Warburg effect, glutaminolysis, and altered fatty acid metabolism).• The role of obesity-induced inflammation and insulin resistance in cancer progression.• The impact of diabetes, hyperglycemia, and metabolic dysfunction on tumorigenesis and cancer progression.• Shared risk factors (e.g., diet, physical inactivity, and smoking) and their contribution to both cancer and metabolic diseases.• Novel therapeutic strategies targeting metabolic pathways in cancer and metabolic diseases.• Integrated approaches combining lifestyle interventions with pharmacological treatments. We welcome a variety of manuscript types, including Original Research, Reviews, Mini-Reviews, and Perspectives, that provide new insights into the mechanisms linking cancer and metabolic diseases. Submissions should aim to advance our understanding of these interconnected conditions and contribute to the development of effective prevention and treatment strategies.
