Metabolic Disorders in Cancer Progression: Insights and Therapeutic Strategies
The intricate relationship between metabolic dysregulation and cancer has emerged as a critical focus within molecular and cellular oncology. Beyond serving as sources of cellular energy, metabolic pathways are now recognized as central regulators of tumor initiation, progression, and therapeutic resistance. Dysfunctions in these tightly regulated pathways -frequently observed in metabolic disorders such as obesity, type 2 diabetes, and hypertension- create a pro-tumorigenic environment that intersects with core oncogenic signaling networks.Recent advances have illuminated significant mechanistic overlap between cancer metabolism and the pathophysiology of metabolic diseases. Aberrant insulin signaling, chronic low-grade inflammation, and altered adipokine profiles are among the key drivers that modulate cellular proliferation, apoptosis, and the tumor microenvironment. These findings are particularly relevant in malignancies such as breast, endometrial, and colorectal cancers, where metabolic disease comorbidities are prevalent and prognostically significant. This Research Topic aims to examine the molecular and cellular mechanisms underlying the link between metabolic disorders and cancer, and to explore how these insights can be leveraged for therapeutic innovation. We welcome contributions employing diverse approaches to dissect the role of metabolic pathways in oncogenesis and tumor progression.Submissions may address, but are not limited to, the following themes:- Molecular mechanisms by which metabolic diseases influence cancer progression- Shared signaling pathways and regulatory networks in metabolic and oncogenic processes- Metabolic reprogramming in cancer cells and the tumor microenvironment- Therapeutic strategies targeting cancer-associated metabolic pathways- Drug discovery and repurposing efforts focused on metabolic disease-related cancersBy fostering interdisciplinary research across oncology and metabolic disease biology, this Research Topic seeks to deepen our understanding of cancer metabolism and advance the development of more effective, mechanism-based therapies.
