Metabolic Regulation and Its Impact on Bone Health in Cancer
Bone is a metabolically active tissue that is continuously remodeled through interactions between osteoblasts, osteoclasts, osteocytes, and systemic metabolic signals. In cancer, these networks may be disrupted by tumor-induced metabolic reprogramming, endocrine dysfunction, chronic inflammation, metabolic conditions, and anticancer therapies. These changes can disturb the calcium–phosphate–PTH–vitamin D axis and contribute to treatment-induced bone loss, musculoskeletal complications, skeletal-related events, and reduced quality of life. Cachexia, sarcopenia, obesity, and insulin resistance may further influence the bone–tumor metabolic interface. Recent advances have highlighted the roles of glucose, lipid, and amino acid metabolism, mitochondrial function, and metabolic signaling in primary bone tumors and cancers that metastasize to bone. Understanding these mechanisms may support strategies that preserve skeletal integrity while improving cancer outcomes. This Research Topic aims to advance understanding of the metabolic mechanisms regulating bone health in cancer. We welcome basic, translational, and clinical studies examining how malignancy, cancer therapies, and endocrine dysfunction influence bone remodeling, mineral metabolism, and skeletal disease progression. Particular emphasis will be placed on the molecular and cellular pathways connecting metabolism, the bone microenvironment, and tumor progression, as well as biomarkers and therapeutic interventions. By integrating these perspectives, this collection will provide new insights into the relationship between metabolism, cancer, and skeletal health. We welcome contributions addressing, but not limited to, the following themes:• Metabolic reprogramming and its effects on bone remodeling and skeletal homeostasis in cancer • Endocrine and metabolic regulation of bone metastasis and the bone tumor microenvironment • Dysregulation of mineral metabolism, including the calcium–phosphate–PTH–vitamin D axis • Cancer treatment–induced bone loss and musculoskeletal complications arising from chemotherapy, radiotherapy, hormonal therapy, and other anticancer treatments • The role of carbohydrate, lipid, amino acid, and mitochondrial metabolism in cancer-associated bone disease • Metabolic interactions among stromal, bone-resident, and malignant cells in the bone microenvironment • Effects of cachexia, sarcopenia, obesity, and insulin resistance on the bone–tumor interface • Biomarkers, imaging approaches, and metabolic therapies for cancer-associated bone disease Topic editors: Chandi C Mandal, Uma Sankar Source: https://www.frontiersin.org/research-topics/84747/metabolic-regulation-and-its-impact-on-bone-health-in-cancer
