Mitochondrial Dynamics and Metabolic Crosstalk in the Tumor Microenvironment: Implications for Immune Modulation and Therapy

The tumor microenvironment (TME) is a dynamic and complex ecosystem composed of cancer cells, immune infiltrates, stromal elements, and diverse metabolic cues. Within this intricate network, mitochondria serve as central regulators of cellular metabolism, signaling, and survival. In cancer, mitochondrial function is profoundly reprogrammed—not only to support the elevated energy and biosynthetic demands of rapidly proliferating tumor cells, but also to influence the behavior, fate, and function of surrounding immune cells. This Research Topic aims to explore the multifaceted roles of mitochondria in mediating metabolic crosstalk between tumors and the immune system. Specifically, we seek to understand how mitochondrial metabolism, nutrient availability, and bioenergetic competition within the TME shape immune surveillance, promote immune evasion, and modulate the outcomes of immunotherapy. Particular emphasis will be placed on how cancer-driven metabolic rewiring alters mitochondrial dynamics— such as fission, fusion, and mitophagy—in both tumor and immune cells, with consequences for immune activation or suppression.Recent advances in immunometabolism and cancer biology have underscored the importance of mitochondrial pathways in controlling not only tumor progression but also the efficacy of immune responses. By dissecting these interactions, this Topic aims to uncover novel mitochondrial-targeted strategies to disrupt the tumor-supportive metabolic landscape and enhance anti-tumor immunity. We welcome the submission of Original Research, Review, Mini Review, and Perspective articles on topics including, but not limited to:• Mitochondrial regulation of immune cell function in the TME• Metabolic reprogramming of tumor-infiltrating lymphocytes• Mitochondrial ROS, dynamics, and mitophagy in immune modulation• Nutrient competition between tumor and immune cells• Mitochondrial mechanisms of immunotherapy resistance• Therapeutic interventions targeting mitochondrial metabolismThis Research Topic invites interdisciplinary perspectives at the intersection of mitochondrial biology, cancer metabolism, and immuno-oncology.
