Mitochondrial Metabolism and Mitochondrial Dysfunction in Hepatotoxic and Metabolic Disease
Mitochondria are central organelles in cellular physiology, orchestrating energy production, redox balance, and cell death pathways. The liver, as a hub for metabolic processes, is especially reliant on mitochondrial function for energy homeostasis, detoxification, and adaptation to metabolic stress. Mitochondrial dysfunction, resulting from genetic, environmental, or pharmacological insults, has emerged as a critical factor in hepatic diseases such as non-alcoholic fatty liver disease (NAFLD), non-alcoholic steatohepatitis (NASH), Metabolic dysfunction-associated steatohepatitis (MASH) , drug-induced liver injury (DILI), and alcoholic liver disease. Recent advances in metabolomics, imaging, and molecular biology have linked mitochondrial bioenergetics, oxidative stress, and organelle crosstalk with hepatocyte survival and death. Nevertheless, central questions remain regarding the mechanistic underpinnings, adaptive responses, and therapeutic targeting of mitochondrial dysfunction in hepatotoxicity and metabolic disorders. This Research Topic seeks to assemble a multidisciplinary collection of cutting-edge research, comprehensive reviews, and short communications to deepen our understanding of mitochondrial metabolism and dysfunction in the context of hepatotoxic and metabolic disease. We welcome original research, brief research reports, (mini-)reviews, methods articles, perspectives, and more that collectively address the roles, regulations, and consequences of mitochondrial alterations in hepatic pathophysiology. By gathering diverse article types from mechanistic studies using novel models to translational research and therapeutic explorations, this collection aims to bridge knowledge gaps and drive innovation towards new diagnostics and interventions.Areas to be covered might include, but are not limited to:• Molecular mechanisms of mitochondrial dysfunction in hepatocytes during toxic and metabolic challenges• Crosstalk between mitochondria and other organelles (ER, lysosomes) in liver cell death and survival• Mitochondrial dynamics (fusion, fission, mitophagy) and their impact on hepatic pathology• Impact of mitochondrial reactive oxygen species (ROS) and redox imbalances in liver disease progression• Role of mitochondrial DNA damage and repair in hepatotoxicity• Metabolomic and proteomic profiling of mitochondrial states in hepatic diseases• Models and methodologies for studying mitochondrial function and dysfunction in liver• Mitochondria-targeted therapies and interventions for hepatotoxic and metabolic liver diseases• Mitochondrial signaling pathways linking metabolism, inflammation, and cell death in the liver Topic editors: Sanda Win, Mrigya Babuta Source: https://www.frontiersin.org/research-topics/76721/mitochondrial-metabolism-and-mitochondrial-dysfunction-in-hepatotoxic-and-metabolic-disease
