Molecular Mechanisms among Cells, Biomaterials, and Microenvironments in Tissue Regeneration
Tissue regeneration is a complex, dynamic biological process involving coordinated interactions between resident and recruited cells, extracellular matrix (ECM), biomaterials, therapeutic agents, and the surrounding biochemical and mechanical microenvironment. Within musculoskeletal, neural, skin, and other soft tissue systems, regeneration extends beyond the mere replacement of damaged tissues to encompass angiogenesis, immune modulation, matrix remodeling, and metabolic adaptation. Recent advances in biomaterials engineering, drug delivery systems, spatial biology, and multi-omics technologies now provide unprecedented insight into these processes at molecular and spatial resolutions. However, despite these developments, the molecular interplays connecting cellular behavior, biomaterial properties, therapeutic stimuli, and microenvironmental cues remain incompletely understood, often limiting our ability to predict or improve regenerative outcomes. There is growing recognition that improved integration of mechanistic molecular evidence with structural and functional assessments could significantly accelerate translational progress in regenerative medicine. This Research Topic aims to bridge the gap between structural regeneration and the underlying molecular mechanisms that drive tissue repair. It seeks to elucidate how cell–cell and cell–matrix communication, biomaterial-mediated signaling, and therapeutic interventions orchestrate repair outcomes across diverse tissue types. Specific objectives include defining the signaling cascades and metabolic pathways underlying effective regeneration, characterizing how local immune activity and matrix organization shape tissue remodeling, and identifying molecular indicators that predict or impair repair efficiency. Contributions are encouraged that link molecular pathways to tissue-level repair, providing mechanistic clarity and experimental validation of biological hypotheses relevant to regenerative processes. The Research Topic focuses on the molecular interactions at the interface of cells, extracellular matrices, biomaterials, therapeutic agents, and local microenvironments during tissue repair and regeneration, with an emphasis on mechanistic understanding rather than purely descriptive outcomes. To gather further insights in this area, we welcome articles addressing, but not limited to, the following themes:o Molecular crosstalk driving cell–cell and cell–matrix communication during tissue regenerationo Biomaterial- and drug-mediated modulation of cellular and tissue responseso Extracellular vesicles, biologics, and other signaling mediators shaping the regenerative milieuo Metabolic, mitochondrial, and redox regulation in repair and remodeling processeso Matrix remodeling dynamics, mechanotransduction, and microenvironmental signalingo Single-cell, spatial, and multi-omics analyses with experimental validation in regenerative modelso Molecular targets and pathways involved in delayed or incomplete tissue repairAppendix:Article types accepted include Original Research, Review, Mini Review, Perspective, Methods, and Brief Research Report.
