Multi-Target Therapeutic Strategies in Neurological Disorders: Beyond Single-Gene Paradigms

Neurological disorders represent a highly complex area within medicine, characterized by multifactorial etiologies involving disruptions across numerous genes, molecular pathways, and brain networks. Traditional drug development has often relied on single-target interventions aimed at correcting isolated molecular defects. However, growing evidence from neurodegenerative, neuropsychiatric, epileptic, and cerebrovascular diseases underscores the limited efficacy and poor sustainability of these approaches, as these conditions rarely stem from singular molecular causes. Recent studies increasingly highlight the need for a paradigm shift—one that acknowledges the web of dysregulated processes and seeks to restore balance at the network level by engaging multiple biological nodes simultaneously.Emerging research demonstrates that multi-target strategies—including rational drug combinations, polypharmacological molecules, multi-component natural products, and interventions designed to recalibrate entire pathways—can yield superior outcomes in restoring neural homeostasis. Advances in systems biology, network pharmacology, integrative omics, and artificial intelligence have enabled researchers to identify key disease modules, actionable hub targets, and synergistic interactions that underpin complex neurological phenotypes. Despite these promising developments, significant knowledge gaps remain regarding optimal target selection, mechanistic synergies, resistance mechanisms, and the translation of network-based findings into clinically actionable therapies. Further, the integration of patient-specific molecular profiling into precision neurology presents both novel opportunities and substantial investigative challenges. This Research Topic aims to advance understanding and application of multi-target therapeutic strategies for neurological diseases by promoting conceptual advances, methodological innovations, and translational research. The objective is to deepen insight into how these multifaceted approaches can overcome current limitations, generate robust and durable effects, and facilitate the transition toward more personalized interventions. Contributions are encouraged to address critical questions including the identification of effective target combinations, the design and validation of polypharmacological compounds, and the real-world implementation of network-guided therapies in neurological populations. To gather further insights into the complexity and management of neurological disorders through multi-target approaches, this Research Topic invites original research, review articles, methodological manuscripts, and translational studies addressing, but not limited to, the following themes:-Systems biology and network pharmacology in neurological disorders-Polypharmacology and rational multi-target drug design-Multi-component natural products and neurotherapeutics-Integrative omics and precision neurology approaches-Artificial intelligence and computational modeling for multi-target discovery-Translational and clinical perspectives of multi-target therapiesThe Research Topic welcomes original research, reviews, methods, and translational studies related to multi-target strategies in neurological disorders.
