Multilevel Endocrine and Genetic Mechanisms of Infertility: HPG Axis Regulation, Gonadal Function, and Metabolic Crosstalk
Infertility research is at the intersection of reproductive endocrinology, genetics, and metabolic biology. Despite decades of investigation, the intricate web of factors governing reproductive success remains only partially illuminated. The dysfunction of the hypothalamic–pituitary–gonadal (HPG) axis, coupled with abnormalities in gonadal function, metabolic derangements such as insulin resistance, and complex genetic contributors, are increasingly recognized as key players in infertility. Current literature has shed light on individual aspects—such as the hormonal regulation of gametogenesis, the influence of metabolic signals on reproductive tissues, and the role of specific gene networks—but the integrative mechanisms that link these multilevel systems are not fully characterized. Recent research has underscored how environmental exposures, endocrine disruptors, and aging further complicate this landscape, heightening the need for a cohesive approach that bridges basic, experimental, and translational lines of inquiry. This Research Topic aims to deepen and unify our mechanistic understanding of infertility by elucidating the endocrine, genetic, and metabolic pathways that underlie its development. The objective is to foster research that not only explores individual components of reproductive dysfunction but also maps the interactions and feedback mechanisms among them. Central questions include: how do disruptions in the HPG axis integrate with genetic mutations and metabolic signals to drive infertility? Can new biomarkers and therapeutic interventions be identified by identifying intersections among these systems? By encouraging interdisciplinary exploration and mechanistic rigor, this topic seeks to advance diagnostic and therapeutic innovation in reproductive endocrinology. The scope of this Research Topic encompasses original and review contributions that address the dynamic and multilevel nature of infertility. Submissions should demonstrate mechanistic insight, experimental rigor, and translational relevance to reproductive endocrinology.
