Nanomedicine in Cancer: From Fundamental Research to Therapeutic Applications

Cancer remains one of the leading causes of morbidity and mortality worldwide, despite significant advances in molecular biology, targeted therapy, and immunotherapy. In recent years, nanomedicine has emerged as a transformative approach in oncology, offering innovative strategies for cancer diagnosis, drug delivery, and therapeutic intervention. Nanomaterials provide powerful tools to study tumor microenvironment dynamics, intracellular signaling pathways, and cancer cell heterogeneity. Engineered nanoparticles can interact selectively with cancer cells, exploit altered membrane properties, and respond to unique tumor microenvironment conditions such as hypoxia, acidic pH, and aberrant vascularization. These properties make nanoplatforms particularly suitable for targeted drug delivery, minimizing systemic toxicity while improving therapeutic efficacy. From a therapeutic perspective, nanomedicine has facilitated the development of multifunctional platforms capable of co-delivering chemotherapeutic agents, nucleic acids, and immunomodulatory compounds. Advances in lipid-based nanoparticles, polymeric carriers, inorganic nanostructures, and biomimetic systems have expanded the possibilities for personalized oncology. Moreover, nanoscale systems are increasingly being explored for gene editing, RNA interference, and CRISPR-based approaches to correct oncogenic mutations or modulate cancer-associated pathways. In parallel, nanodiagnostics and theranostic platforms are enabling real-time monitoring of tumor progression and treatment response. The integration of imaging agents within nanocarriers enhances early detection and supports precision medicine strategies. Importantly, understanding nanoparticle–cell interactions, cellular uptake mechanisms, endosomal escape, and intracellular trafficking remains central to optimizing translational outcomes. This Research Topic aims to bridge fundamental discoveries in cancer cell biology with translational nanomedicine strategies, fostering interdisciplinary dialogue between basic scientists and clinicians.
